RESUMO: As plantas desde muito tempo são utilizadas na medicina popular, substâncias purificadas e isoladas a partir da matéria-prima vegetal, com estrutura química definida e atividade farmacológica, são denominadas fitofármacos e podem ser usados como ativos em medicamentos com propriedade profilática, paliativa ou
curativa. Têm-se aumentado o interesse em explorar propriedades bioquímicas
dos metabólicos secundários (MS), os flavonoides são exemplos desses
metabólitos e possuem inúmeras propriedades como antioxidantes,
antiinflamatórias, anticancerígenas, antivirais e analgésicas. Além de serem
responsáveis pela ação farmacológica da planta, alguns desses MS, também
podem ser tóxicos, assim testes de toxicidade são de extrema importância e
nesse contexto o estudo teve objetivo de avaliar a toxicidade da esterubina em
modelos in sílico, in vitro e in vivo. Para avaliação in sílico foi utilizada a
plataforma ADMETLab 3.0, onde o smile da substância foi adicionado e a partir
disso dados físico-químicos e de toxicidade foram coletados. Para avaliação in
vitro da toxicidade foi realizado o teste de hemólise, onde foram utilizados
eritrócitos de Canis lupus familiaris, com análise em triplicata as amostras foram
centrifugadas a 3000 rpm por 5 minutos e quantificada a presença de
hemoglobina, na análise in vitro da toxicidade pelo método MTT foram utilizadas
linhagens celulares L929 (fibroblastos de camundongos) e B16F10 (melanoma
murino) e avaliados quanto a viabilidade celular. Para realização dos testes in
vivo foram utilizadas larvas de Zophobas morio a análise foi feita em duplicata
com diluição da substância para obtenção das doses de 3, 30 e 300 mg/kg, que
são doses já preconizadas para o teste de toxicidade, e foram administradas na
hemocele do tenébrio sendo verificadas a melanização e sobrevivência. A
previsão de toxicidade pelo ADMETlab 3.0 trouxe resultados promissores,
mostrando a molécula de esterubina com valores favoráveis de propriedades
como genotoxicidade, mutagenicidade no teste AMES, irritação ocular, e valores
com toxicidade média que não impedem seu uso, mas merecem atenção
especial como carcinogenicidade. No teste de hemólise os resultados foram
positivos apresentando porcentagem menor que 10%. Na avaliação de
citotoxicidade pelo método MTT, a molécula teve eficácia em reduzir a
viabilidade quando comparadas ao controle negativo assim como também o
valor de IC50 de 15,07 μg/mL (12,55 a 18,10 μg/mL) em células B16F10,
mostrando também um bom potencial em reduzir a viabilidade nessa linhagem
celular. No teste in vivo com larvas de Zophobas morio apresentou boa taxa de
sobrevivência em doses maiores de 30 e 300 mg/kg, demonstrando segurança.
Com base nos resultados a esterubina se mostra uma molécula promissora com
bom perfil de segurança demonstrado nos testes in sílico, in vitro e in vivo se
mostrando segura para futuros testes in vivo que possam vir a serem realizados
para evidenciar suas possíveis atividades farmacológicas. ABSTRACT: Plants have long been used in folk medicine. Purified substances isolated from
plant raw materials, with a defined chemical structure and pharmacological
activity, are called phytopharmaceuticals and can be used as active ingredients
in medicines with prophylactic, palliative or curative properties. There has been
increasing interest in exploring the biochemical properties of secondary
metabolites (SM). Flavonoids are examples of these metabolites and have
numerous properties such as antioxidants, anti-inflammatory, anticancer, antiviral
and analgesics. In addition to being responsible for the pharmacological action of
the plant, some of these SM can also be toxic, so toxicity tests are extremely
important and in this context the study aimed to evaluate the toxicity of sterubin
in in silico, in vitro and in vivo models. For in silico evaluation, the ADMETLab 3.0
platform was used, where the substance's smile was added and from this,
physicochemical and toxicity data were collected. For in vitro toxicity evaluation,
the hemolysis test was performed, where Canis lupus familiaris erythrocytes were
used, with analysis in triplicate. The samples were centrifuged at 3000 rpm for 5
minutes and the presence of hemoglobin was quantified. In the in vitro toxicity
analysis by the MTT method, L929 (mouse fibroblasts) and B16F10 (murine
melanoma) cell lines were used and evaluated for cell viability. To perform the in
vivo tests, Zophobas morio larvae were used. The analysis was performed in
duplicate with dilution of the substance to obtain doses of 3, 30 and 300 mg/kg,
which are doses already recommended for the toxicity test, and were
administered in the hemocoel of the tenebrio, verifying melanization and survival.
The toxicity prediction by ADMETlab 3.0 brought promising results, showing the
sterubin molecule with favorable values of properties such as genotoxicity,
mutagenicity in the AMES test, eye irritation, and values with medium toxicity that
do not prevent its use, but deserve special attention as carcinogenicity. In the
hemolysis test, the results were positive, with a percentage lower than 10%. In
the cytotoxicity evaluation by the MTT method, the molecule was effective in
reducing viability when compared to the negative control, as well as the IC50 value
of 15.07 μg/mL (12.55 to 18.10 μg/mL) in B16F10 cells, also showing good
potential in reducing viability in this cell line. In the in vivo test with Zophobas
morio larvae, it showed a good survival rate at doses higher than 30 and 300
mg/kg, demonstrating safety. Based on the results, sterubin is a promising
molecule with a good safety profile demonstrated in in silico, in vitro and in vivo
tests, proving safe for future in vivo tests that may be performed to demonstrate
its possible pharmacological activities. |
