RESUMO: O presente estudo investigou os efeitos antioxidantes e vasoprotetores de uma manteiga industrializada obtida das sementes de Platonia insignis Mart. (MIB) em modelo experimental de estresse oxidativo induzido por etanol, visando sua potencial aplicação no tratamento de doenças cardiovasculares. Inicialmente, foram revisados os mecanismos do estresse oxidativo na fisiopatologia da hipertensão arterial sistêmica, destacando o papel das espécies reativas de oxigênio e nitrogênio na disfunção endotelial e os efeitos deletérios do etanol sobre o sistema cardiovascular, bem como o potencial de compostos antioxidantes em restabelecer o equilíbrio redox. Em seguida, por meio de revisão sistemática baseada no protocolo PRISMA, foi realizada uma análise da literatura científica e de patentes publicada entre 2001 e 2020,
evidenciando o uso de plantas medicinais no manejo da hipertensão e os mecanismos de ação dos seus princípios ativos. O estudo também abordou o potencial terapêutico de P. insignis, espécie amazônica da família Clusiaceae, reconhecida por seus metabólitos bioativos com propriedades antioxidantes. Revisões fitoquímicas indicaram que diferentes partes da planta e compostos isolados, como a benzofenona garcinielliptona FC e a xantona γ-mangostina, apresentam capacidade de inibir a peroxidação lipídica, sequestrar radicais livres e modular vias celulares associadas ao estresse oxidativo, incluindo NRF2 e SIRT1, além de inibir enzimas pró-oxidantes como xantina oxidase e NADPH oxidase. Por fim, a investigação experimental in vivo com ratas Wistar demonstrou que a MIB, em doses de 25 e 50 mg/kg, modulou marcadores bioquímicos e antioxidantes, atenuou alterações histopatológicas hepáticas e
cardíacas induzidas pelo etanol e melhorou a reatividade vascular, promovendo vasoconstrição adequada e vasorrelaxamento endotelial. Os resultados evidenciam que a manteiga de P. insignis possui potencial cardioprotetor frente ao estresse oxidativo e à disfunção vascular induzidos pelo etanol, reforçando a necessidade de estudos adicionais para elucidar seus mecanismos de ação e consolidar seu uso terapêutico. ABSTRACT: This study investigated the antioxidant and vasoprotective effects of an industrially produced butter obtained from the seeds of Platonia insignis Mart. (MIB) in an experimental model of ethanol-induced oxidative stress, aiming at its potential application in the treatment of cardiovascular diseases. Initially, the mechanisms of oxidative stress in the pathophysiology of systemic arterial hypertension were reviewed, highlighting the role of reactive oxygen and nitrogen species in endothelial dysfunction and the deleterious effects of ethanol on the cardiovascular system, as well as the potential of antioxidant compounds to restore redox balance. Subsequently, through a systematic review based on the PRISMA protocol, an analysis
of the scientific literature and patents published between 2001 and 2020 was carried out, highlighting the use of medicinal plants in the management of hypertension and the mechanisms of action of their active principles. The study also addressed the therapeutic potential of P. insignis, an Amazonian species of the Clusiaceae family, recognized for its bioactive metabolites with antioxidant properties. Phytochemical reviews have indicated that different parts of the plant and isolated compounds, such as the benzophenone garcinielliptone FC and the xanthone γ-mangostin, have the ability to inhibit lipid peroxidation, scavenge free radicals, and modulate cellular pathways associated with oxidative stress, including NRF2 and SIRT1, as well as inhibit pro-oxidant enzymes such as xanthine oxidase and NADPH oxidase. Finally, in vivo experimental investigation with Wistar rats demonstrated that MIB, at doses of 25 and
50 mg/kg, modulated biochemical and antioxidant markers, attenuated hepatic and cardiac histopathological alterations induced by ethanol, and improved vascular reactivity, promoting adequate vasoconstriction and endothelial vasorelaxation. The results show that P. insignis butter has cardioprotective potential against oxidative stress and vascular dysfunction induced by ethanol, reinforcing the need for further studies to elucidate its mechanisms of action and consolidate its therapeutic use. |
