Abstract:
RESUMO: A leishmaniose visceral permanece um agravo zoonótico relevante, e estimativas sorológicas
em cães podem variar conforme o desempenho dos ensaios diagnósticos. Este estudo teve
como objetivo estimar e comparar a soroprevalência para Leishmania infantum em cães
domiciliados e encoleirados com coleiras impregnadas com deltametrina 4% nos municípios
de Timon e Caxias (Maranhão, Brasil), por meio dos ensaios EIE-LVC Bio-
Manguinhos/Fiocruz e ELISA rK28. Foi conduzido um estudo transversal com cães
domiciliados inscritos no programa de encoleiramento em Timon (n = 194) e Caxias (n =
211). As amostras de soro foram analisadas pelo ELISA padrão (Bio-Manguinhos/Fiocruz) e
por ELISA utilizando a proteína recombinante K28. A comparação entre os testes dentro de
cada município foi realizada pelo teste de McNemar (dados pareados), e a comparação dos
mesmos testes entre municípios pelo teste do qui-quadrado, considerando p<0,05. A
concordância entre os testes foi avaliada pelo coeficiente Kappa. Os resultados de
soroprevalência foram apresentados em percentagem (%). No ELISA padrão e no ELISA
rK28, a soropositividade foi de 9,8% (19/194) e 2,6% (5/194) em Timon, e 6,6% (14/211) e
4,3% (9/211) em Caxias, respectivamente. Não houve diferença significativa entre
municípios para os diferentes testes. Entretanto, houve diferença significativa entre os testes
dentro de cada município, com maior positividade no teste Bio-Manguinhos em relação ao
rK28. Conclui-se que a soroprevalência foi maior no ELISA padrão do que no ELISA rK28,
em Timon e Caxias, sem diferença significativa entre municípios para cada ensaio. A
concordância fraca entre os testes sugere que não são intercambiáveis. ABSTRACT: Visceral leishmaniasis remains a relevant zoonotic disease, and serological estimates in dogs
may vary according to the performance of diagnostic assays. This study aimed to estimate and
compare the seroprevalence of Leishmania infantum in household dogs wearing 4%
deltamethrin-impregnated collars in the municipalities of Timon and Caxias (Maranhão,
Brazil), using the EIE-LVC Bio-Manguinhos/Fiocruz assay and the rK28 ELISA. A cross-
sectional study was conducted with household dogs enrolled in the collaring program in
Timon (n = 194) and Caxias (n = 211). Serum samples were tested using the standard ELISA
(Bio-Manguinhos/Fiocruz) and an ELISA based on the recombinant K28 protein. Within each
municipality, paired comparisons between tests were performed using McNemar’s test, and
comparisons of the same tests between municipalities were performed using the chi-square
test, considering p < 0.05. Agreement between tests was assessed using the Kappa coefficient.
Seroprevalence results were expressed as percentages (%). In the standard ELISA and the
rK28 ELISA, seropositivity was 9.8% (19/194) and 2.6% (5/194) in Timon, and 6.6%
(14/211) and 4.3% (9/211) in Caxias, respectively. There was no significant difference
between municipalities for either test. However, there was a significant difference between
tests within each municipality, with higher positivity in the Bio-Manguinhos assay than in the
rK28 ELISA. In conclusion, seroprevalence was higher with the standard ELISA than with
the rK28 ELISA in Timon and Caxias, with no significant between-municipality differences
for each assay. The weak agreement between tests suggests that they are not interchangeable.
