<title>AVALIAÇÃO ELETROCARDIOGRÁFICA DA SOBRECARGA ATRIAL ESQUERDA EM CÃES: correlação com o estadiamento  ACVIM e parâmetros ecocardiográficos</title>

RESUMO: A doença mixomatosa valvar mitral (DMVM) requer identificação acurada do aumento&#13;
atrial esquerdo (AAE) para orientar o estadiamento e a terapêutica. Embora a eletrocardiografia&#13;
(ECG) seja amplamente disponível e de baixo custo, sua acurácia isolada para AAE é limitada.&#13;
Assim, este estudo investigou a derivação e a validação interna de um algoritmo de triagem&#13;
eletrocardiográfico multifatorial, combinando a duração da onda P na derivação II e o índice de&#13;
Morris, com o objetivo de maximizar a sensibilidade para detectar AAE clinicamente relevante,&#13;
definido por razão átrio esquerdo/aorta (AE/Ao) ≥ 1,6. Em um estudo retrospectivo, foram&#13;
incluídos 150 cães nos estágios A a C da ACVIM. O AE/Ao ≥ 1,6 ao ecocardiograma foi&#13;
adotado como padrão de referência para AAE. As mensurações eletrocardiográficas foram&#13;
avaliadas quanto à reprodutibilidade interobservador, e o algoritmo foi desenvolvido e testado&#13;
por validação interna com cross-validation e limiares otimizados, com exploração de utilidade&#13;
clínica por análise de curva de decisão (DCA). O algoritmo apresentou área sob a curva (AUC)&#13;
de 0,729. Considerando-se as categorias de risco Alto ou Intermediário como limiar de triagem,&#13;
obteve-se sensibilidade de 88,5% e valor preditivo negativo (VPN) de 96,6%. A DCA indicou&#13;
maior benefício líquido da estratificação de risco em comparação a marcadores&#13;
eletrocardiográficos isolados em ampla faixa de limiares de decisão clínica. Os achados devem&#13;
ser interpretados com cautela devido ao risco de otimismo analítico inerente à validação interna&#13;
&#13;
e ao uso do AE/Ao em vez do índice de volume atrial esquerdo (LAVi) como principal padrão-&#13;
ouro. Conclui-se que o algoritmo de triagem por ECG demonstrou segurança clínica,&#13;
&#13;
permitindo excluir AAE clinicamente relevante com razoável confiança e orientar o&#13;
encaminhamento para ecocardiografia em cães classificados como risco intermediário ou alto. ABSTRACT: Myxomatous mitral valve disease (MMVD) requires accurate identification of left atrial&#13;
enlargement (LAE) to guide clinical staging and therapy. Because surface electrocardiography&#13;
(ECG) is widely available and low-cost but has limited standalone accuracy for LAE, this study&#13;
investigated whether a multifactorial ECG-based screening algorithm could improve detection&#13;
of clinically relevant LAE. In a retrospective design, 150 dogs across ACVIM stages A to C&#13;
were evaluated. Echocardiographic LAE was defined using left atrial-to-aortic root ratio&#13;
(LA/Ao) ≥ 1.6 as the reference standard. Electrocardiographic measurements were tested for&#13;
interobserver reproducibility, and an algorithm combining P-wave duration in lead II with the&#13;
Morris index was derived and internally validated using cross-validation with optimized&#13;
thresholds; clinical utility was further explored with decision curve analysis (DCA). The&#13;
algorithm achieved an area under the curve (AUC) of 0.729. When Intermediate- or High-Risk&#13;
categories were used as the screening threshold, sensitivity was 88.5% and negative predictive&#13;
value (NPV) was 96.6%, supporting its use to rule out clinically relevant LAE with reasonable&#13;
confidence. DCA indicated a greater net benefit for risk stratification than for isolated ECG&#13;
markers across a broad range of decision thresholds. Findings should be interpreted with caution&#13;
due to potential analytical optimism from internal validation and the use of LA/Ao rather than&#13;
left atrial volume index (LAVi) as the primary reference. Overall, the ECG-based screening&#13;
algorithm may assist referral decisions for echocardiography, particularly for dogs classified as&#13;
intermediate or high risk.

RESUMO: A leishmaniose visceral permanece um agravo zoonótico relevante, e estimativas sorológicas&#13;
em cães podem variar conforme o desempenho dos ensaios diagnósticos. Este estudo teve&#13;
como objetivo estimar e comparar a soroprevalência para Leishmania infantum em cães&#13;
domiciliados e encoleirados com coleiras impregnadas com deltametrina 4% nos municípios&#13;
&#13;
de Timon e Caxias (Maranhão, Brasil), por meio dos ensaios EIE-LVC Bio-&#13;
Manguinhos/Fiocruz e ELISA rK28. Foi conduzido um estudo transversal com cães&#13;
&#13;
domiciliados inscritos no programa de encoleiramento em Timon (n = 194) e Caxias (n =&#13;
211). As amostras de soro foram analisadas pelo ELISA padrão (Bio-Manguinhos/Fiocruz) e&#13;
por ELISA utilizando a proteína recombinante K28. A comparação entre os testes dentro de&#13;
cada município foi realizada pelo teste de McNemar (dados pareados), e a comparação dos&#13;
mesmos testes entre municípios pelo teste do qui-quadrado, considerando p&lt;0,05. A&#13;
concordância entre os testes foi avaliada pelo coeficiente Kappa. Os resultados de&#13;
soroprevalência foram apresentados em percentagem (%). No ELISA padrão e no ELISA&#13;
rK28, a soropositividade foi de 9,8% (19/194) e 2,6% (5/194) em Timon, e 6,6% (14/211) e&#13;
4,3% (9/211) em Caxias, respectivamente. Não houve diferença significativa entre&#13;
municípios para os diferentes testes. Entretanto, houve diferença significativa entre os testes&#13;
dentro de cada município, com maior positividade no teste Bio-Manguinhos em relação ao&#13;
rK28. Conclui-se que a soroprevalência foi maior no ELISA padrão do que no ELISA rK28,&#13;
em Timon e Caxias, sem diferença significativa entre municípios para cada ensaio. A&#13;
concordância fraca entre os testes sugere que não são intercambiáveis. ABSTRACT: Visceral leishmaniasis remains a relevant zoonotic disease, and serological estimates in dogs&#13;
may vary according to the performance of diagnostic assays. This study aimed to estimate and&#13;
compare the seroprevalence of Leishmania infantum in household dogs wearing 4%&#13;
deltamethrin-impregnated collars in the municipalities of Timon and Caxias (Maranhão,&#13;
&#13;
Brazil), using the EIE-LVC Bio-Manguinhos/Fiocruz assay and the rK28 ELISA. A cross-&#13;
sectional study was conducted with household dogs enrolled in the collaring program in&#13;
&#13;
Timon (n = 194) and Caxias (n = 211). Serum samples were tested using the standard ELISA&#13;
(Bio-Manguinhos/Fiocruz) and an ELISA based on the recombinant K28 protein. Within each&#13;
municipality, paired comparisons between tests were performed using McNemar’s test, and&#13;
comparisons of the same tests between municipalities were performed using the chi-square&#13;
test, considering p &lt; 0.05. Agreement between tests was assessed using the Kappa coefficient.&#13;
Seroprevalence results were expressed as percentages (%). In the standard ELISA and the&#13;
rK28 ELISA, seropositivity was 9.8% (19/194) and 2.6% (5/194) in Timon, and 6.6%&#13;
(14/211) and 4.3% (9/211) in Caxias, respectively. There was no significant difference&#13;
between municipalities for either test. However, there was a significant difference between&#13;
tests within each municipality, with higher positivity in the Bio-Manguinhos assay than in the&#13;
rK28 ELISA. In conclusion, seroprevalence was higher with the standard ELISA than with&#13;
the rK28 ELISA in Timon and Caxias, with no significant between-municipality differences&#13;
for each assay. The weak agreement between tests suggests that they are not interchangeable.

<summary type="text">AVALIAÇÃO DA FORMA DE FORNECIMENTO DE SILAGEM NA DIETA SOBRE A COMPOSIÇÃO TECIDUAL E QUALIDADE DA CARNE DE CORDEIROS TERMINADOS EM CONFINAMENTO

RESUMO: Com a realização desta pesquisa objetivou-se avaliar a influência do tipo de silagem&#13;
sobre a composição tecidual, características físico-químicas e o perfil de ácidos&#13;
graxos da carne de ovinos terminados em confinamento. No Cap. I foi feita uma&#13;
revisão integrativa, em que as buscas para a seleção dos artigos foram realizadas&#13;
nas bases: SCOPUS, Web of Science, Wiley Online Library e Science direct. Para&#13;
tanto, usou da estratégia de pesquisa a mnemônica PVO (population, variable of&#13;
interest, outcome) para então definir a pergunta norteadora da pesquisa: “Quais as&#13;
principais silagens usadas na terminação de ovinos?”. No Cap. II, foi conduzido&#13;
um experimento com 32 cordeiros mestiços, machos, com peso inicial médio de&#13;
±19,6 kg terminados em confinamento, alimentados com silagem de ração em&#13;
mistura total (SRMT) contendo pornunça (Manihot sp.). Os animais foram&#13;
distribuídos em um delineamento de blocos completos casualizados, em arranjo&#13;
fatorial 2 × 2, sendo duas formas de fornecimento (FF; SRMT e RMT) e duas&#13;
relações volumosos:concentrado (V:C 40:60 e 50:50). Os dados foram analisados&#13;
usando modelos mistos, sendo os tratamentos e suas interações considerados&#13;
efeitos fixos e os efeitos de bloco e o erro experimental, efeitos aleatórios. As&#13;
médias foram comparadas pelo teste ajustado de Tukey, sendo consideradas&#13;
diferentes quando P ≤ 0,05. A revisão integrativa revelou que o Brasil foi o país que&#13;
mais desenvolveu estudos sobre o tema, sendo a silagem da planta de milho a mais&#13;
utilizada como fonte de volumoso na alimentação de ovinos confinados. De modo&#13;
geral, o uso de diferentes silagens nas dietas exerce influência direta sobre a&#13;
qualidade da carne de cordeiros confinados, a composição tecidual,&#13;
características físico-químicas e ainda o perfil lipídico. Em relação ao Cap. II,&#13;
as dietas ofertadas na forma de RMT proporcionaram maior peso da perna (P =&#13;
0,02), independentemente da relação V:C. Observou-se que a dieta com&#13;
SRMT40:60 apresentou maior proporção de ossos (O) em comparação à&#13;
RMT40:60. As proporções dos demais tecidos (M = músculo; G = gordura e outros&#13;
tecidos) não foram influenciadas (P &gt; 0,05) pelas dietas. O índice de&#13;
musculosidade, bem como as relações M:O e M:G também não apresentaram&#13;
diferença significativa (P &gt; 0,05). As dietas não influenciaram a composição&#13;
centesimal da carne (P &gt; 0,05). Os maiores valores para L* (luminosidade) e perdas&#13;
por cocção (PPC) foram observados na relação 50:50 em comparação à relação&#13;
40:60. As demais características físicas não foram influenciadas pelas dietas (P &gt;&#13;
0,05). Os animais alimentados com SRMT, independente da relação V:C,&#13;
apresentaram carne com maior concentração de C15:1 e C18:3 n-3 comparados&#13;
aos alimentados com RMT. A relação V:C de 40:60 proporcionou carne com maior&#13;
concentração de C20:2. Os demais ácidos graxos não foram influenciados pelas&#13;
dietas (P &gt; 0,05). Assim, com base nos resultados conclui-se que a Pornunça pode&#13;
ser utilizada com segurança como fonte de volumoso tanto na forma de SRMT&#13;
quanto na forma de RMT, nas relações V:C de 40:60 ou 50:50, sem prejuízo à&#13;
qualidade da carne. ABSTRACT: The objective of this research was to evaluate the influence of silage type on the&#13;
tissue composition, physicochemical characteristics, and fatty acid profile of lamb&#13;
meat from animals finished in confinement. In Chapter I, an integrative review was&#13;
carried out, in which the searches for article selection were conducted in the&#13;
databases: SCOPUS, Web of Science, Wiley Online Library, and ScienceDirect.&#13;
The mnemonic PVO (population, variable of interest, outcome) research strategy&#13;
was used to define the guiding question of the study: “What are the main silages&#13;
used in the finishing of sheep?”. In Chapter II, an experiment was conducted with&#13;
32 crossbred male lambs, with an initial average weight of ±19.6 kg, finished in&#13;
confinement and fed with total mixed ration silage (TMRS) containing pornunça&#13;
(Manihot sp.). The animals were distributed in a randomized complete block design,&#13;
arranged in a 2 × 2 factorial scheme, with two feeding forms (FF; TMRS and TMR)&#13;
and two forage-to-concentrate ratios (F:C 40:60 and 50:50). Data were analyzed&#13;
using mixed models, with treatments and their interactions considered as fixed&#13;
effects, and block and experimental error as random effects. Means were compared&#13;
using Tukey’s adjusted test, and differences were considered significant when P ≤&#13;
0.05. The integrative review revealed that Brazil was the country with the highest&#13;
number of studies on the topic, and corn plant silage was the most used roughage&#13;
source in confined sheep feeding. Overall, the use of different silages in the diet&#13;
exerts a direct influence on lamb meat quality, tissue composition, physicochemical&#13;
characteristics, and lipid profile. Regarding Chapter II, the diets supplied in the form&#13;
of TMR resulted in higher leg weight (P = 0.02), regardless of the F:C ratio. The diet&#13;
with TMRS40:60 showed a higher proportion of bone (B) compared to TMR40:60.&#13;
The proportions of the other tissues (M = muscle; F = fat; and other tissues) were&#13;
not affected (P &gt; 0.05) by the diets. The muscularity index, as well as the M:B and&#13;
M:F ratios, also showed no significant differences (P &gt; 0.05). The diets did not&#13;
influence the proximate composition of the meat (P &gt; 0.05). Higher values for L*&#13;
(lightness) and cooking losses (CL) were observed for the 50:50 ratio compared to&#13;
40:60. The other physical characteristics were not affected by the diets (P &gt; 0.05).&#13;
Animals fed with TMRS, regardless of the F:C ratio, showed meat with higher&#13;
concentrations of C15:1 and C18:3 n-3 compared to those fed with TMR. The 40:60&#13;
F:C ratio resulted in meat with a higher concentration of C20:2. The other fatty acids&#13;
were not affected by the diets (P &gt; 0.05). Thus, based on the results, it can be&#13;
concluded that pornunça can be safely used as a roughage source, either in the&#13;
TMRS or TMR form, with F:C ratios of 40:60 or 50:50, without compromising meat&#13;
quality.

<summary type="text">POTENCIAL BIOTECNOLOGICO DE PROTEASES EXTRAIDAS DA CASCA DO FRUTO DA CASTANHA-DO-GURGUEIA (DIPTERYX LACUNIFERA DUCKE) E SUA APLICAÇÃO EM RAÇÕES PARA FRANGOS DE CORTE

RESUMO: A castanheira-de-gurguéia (Dipteryx lacunifera Ducke), popularmente conhecida como "castanheira de burro" ou "fava de morcego", é uma espécie nativa da região Nordeste do Brasil, reconhecida principalmente pelas suas amêndoas, que contêm elevados níveis de lipídios e proteínas. Contudo, o potencial biotecnológico de seus resíduos, como a casca dos frutos, ainda é pouco explorado. A casca da castanheira pode ser uma fonte promissora para a extração de enzimas, como as proteases, que podem ter aplicações no setor agropecuário, especialmente na alimentação animal. A avicultura de corte, uma importante atividade econômica para o Brasil, enfrenta desafios relacionados ao estresse térmico, que prejudica o bem-estar das aves e afeta seu desempenho produtivo. Como os frangos são animais homeotérmicos, têm dificuldades em&#13;
regular sua temperatura corporal em condições de calor extremo, o que pode resultar em&#13;
impactos negativos na produtividade. Portanto, explorar alternativas nutricionais que ajudem a mitigar os efeitos do estresse térmico é uma área de grande interesse para a avicultura. Este estudo teve como objetivo avaliar o potencial biotecnológico de proteases extraídas da casca do fruto da castanheira-do-gurguéia (Dipteryx lacunifera Ducke) e sua aplicação em rações para frangos de corte. Além disso, buscou-se verificar os efeitos da adição de extrato bruto e purificado dessas proteases na dieta de pintos de corte durante a fase de 1 a 7 dias de idade, sob condições de estresse térmico. O extrato bruto de proteases foi obtido através da maceração da casca dos frutos de D. lacunifera, seguido de extração com tampão fosfato de sódio 0,1 M a pH 8, agitação por 120 minutos e centrifugação. A atividade das proteases e a quantificação proteica foi avaliada, assim como a caracterização bioquímica da enzima que incluiu a determinação do pH ótimo, temperatura ótima e suas estabilidade, assim como a influência de íons e surfactantes sobre a atividade enzimática da protease. Foi feita ainda a avaliação da toxicidade do extrato frente a Artemia salina e a estabilidade da enzima sobre rações comerciais&#13;
para frangos de corte. A purificação das enzimas foi realizada utilizando o sistema de duas fases aquosas (PEG/Sal) através de um planejamento fatorial 2⁴. Para a avaliação da aplicação da enzima para analise in vivo, 150 pintos da linhagem Cobb foram distribuídos em um experimento totalmente casualizado, com três tratamentos: grupo controle (sem enzima), grupo com enzima extraída do extrato bruto (300 unidades/g de ração) e grupo com enzima purificada (6,6 g/kg de ração). Os pintos foram expostos a condições de estresse térmico durante a fase de 1 a 7 dias de idade. As proteases extraídas apresentaram uma atividade de 30,66 U/mL e uma atividade específica de 66,65 U/mg. A caracterização bioquímica revelou um pH ótimo de 11, parcialmente estável após 24 horas e uma temperatura ótima de 50°C, com estabilidade por até 90 minutos. A adição de Mg²⁺ aumentou a atividade enzimática em 11%, enquanto os demais&#13;
íons reduziram a atividade. O extrato mostrou-se estável em rações peletizadas e fareladas e não apresentou toxicidade para Artemia salina. A purificação utilizando o sistema de duas fases aquosas resultou em uma recuperação de 204,22% da atividade enzimática, com um fator de purificação de 8,6. Quanto ao desempenho das aves, os resultados mostraram que, após 7 dias de estresse térmico, os pintos que receberam o extrato purificado apresentaram ganho de peso significativamente maior em comparação aos grupos controle e com extrato bruto. As proteases extraídas da casca do fruto da Dipteryx lacunifera Ducke demonstraram características bioquímicas favoráveis, elevada estabilidade em rações e ausência de toxicidade, indicando seu potencial como aditivo enzimático sustentável na nutrição animal. Além disso, a suplementação com esse extrato purificado melhorou significativamente o ganho de peso dos frangos de corte, mesmo sob condições severas de estresse térmico, destacando o valor dessa alternativa na avicultura. Este estudo sugere que a utilização dessas proteases pode ser uma estratégia promissora para melhorar os parâmetros produtivos dos frangos, como ganho de peso e conversão alimentar, especialmente quando combinada com medidas ambientais adequadas para reduzir os efeitos do estresse térmico. ABSTRACT: The castanheira-de-gurguéia (Dipteryx lacunifera Ducke), popularly known as "castanheira de&#13;
burro" or "fava de morcego," is a species native to the Northeast region of Brazil, primarily&#13;
recognized for its almonds, which contain high levels of lipids and proteins. However, the&#13;
biotechnological potential of its residues, such as the fruit peel, remains largely unexplored.&#13;
The peel of the castanheira may be a promising source for the extraction of enzymes, such as&#13;

Broiler poultry farming, an important economic activity in Brazil, faces challenges related to&#13;
thermal stress, which harms the well-being of the birds and affects their productive&#13;
performance. Since chickens are homeothermic animals, they have difficulty regulating their&#13;
body temperature in extreme heat conditions, which can result in negative impacts on&#13;
productivity. Therefore, exploring nutritional alternatives to help mitigate the effects of thermal&#13;
stress is an area of great interest for poultry farming. This study aimed to evaluate the&#13;
biotechnological potential of proteases extracted from the fruit peel of castanheira-de-gurguéia&#13;
(Dipteryx lacunifera Ducke) and their application in broiler poultry feed. Additionally, the&#13;
study sought to assess the effects of adding both crude and purified extracts of these proteases&#13;
to the diet of day-old chicks (1-7 days of age) under thermal stress conditions. The crude&#13;
protease extract was obtained by macerating the fruit peel of D. lacunifera, followed by&#13;
extraction with 0.1 M sodium phosphate buffer at pH 8, agitation for 120 minutes, and&#13;
centrifugation. The protease activity and protein quantification were evaluated, as well as the&#13;
biochemical characterization of the enzyme, which included determining the optimal pH,&#13;
optimal temperature, and enzyme stability, along with the influence of ions and surfactants on&#13;
enzyme activity. The toxicity of the extract was assessed using Artemia salina, and the enzyme&#13;
stability in commercial broiler feed was also evaluated. Enzyme purification was carried out&#13;
using a two-phase aqueous system (PEG/Sal) through a 2⁴ factorial design. For the in vivo&#13;
application assessment, 150 Cobb chicks were distributed in a completely randomized&#13;
experiment with three treatments: a control group (without enzyme), a group with enzyme&#13;
extracted from the crude extract (300 units/g of feed), and a group with purified enzyme (6.6&#13;
g/kg of feed). The chicks were exposed to thermal stress conditions during the first 7 days of&#13;
age. The extracted proteases presented an activity of 30.66 U/mL and a specific activity of 66.65&#13;
U/mg. The biochemical characterization revealed an optimal pH of 11, which was partially&#13;
stable after 24 hours, and an optimal temperature of 50°C, with stability for up to 90 minutes.&#13;
The addition of Mg²⁺ increased enzyme activity by 11%, while other ions reduced the activity.&#13;
The extract was stable in pelleted and crumbled feed and showed no toxicity to Artemia salina.&#13;
The purification using the two-phase aqueous system resulted in a recovery of 204.22% of&#13;
enzyme activity, with a purification factor of 8.6. Regarding bird performance, the results&#13;
showed that after 7 days of thermal stress, the chicks that received the purified extract had a&#13;
significantly higher weight gain compared to the control and crude extract groups. Proteases&#13;
extracted from the fruit peel of Dipteryx lacunifera Ducke demonstrated favorable biochemical&#13;
characteristics, high stability in feed, and no toxicity, indicating their potential as a sustainable&#13;
enzymatic additive in animal nutrition. Moreover, supplementation with this purified extract&#13;
significantly improved the weight gain of broiler chickens, even under severe thermal stress&#13;
conditions, highlighting the value of this alternative in poultry farming. This study suggests that&#13;
the use of these proteases can be a promising strategy to improve productive parameters in&#13;
broilers, such as weight gain and feed conversion, especially when combined with appropriate&#13;
environmental measures to reduce the effects of thermal stress
