RESUMO: Em regiões semiáridas, plantas tolerantes à seca, como a palma forrageira (family Cactaceae), são essenciais para sustentar a produção de alimentos para humanos e animais. No entanto, secas prolongadas, intensificadas pelas mudanças climáticas, podem comprometer o crescimento dessa planta. Alguns táxons bacterianos, como Bacillus e Paenibacillus, são conhecidos por colonizar a rizosfera e promover o crescimento das plantas (BPCP), além de atenuar os efeitos do estresse hídrico. No entanto, pouco se sabe sobre os efeitos da inoculação em cactáceas na biomassa microbiana, na atividade enzimática e no microbioma da rizosfera&#13;
de plantas sob as condições edafoclimáticas do semiárido. Nesse contexto, o objetivo dessa Tese de doutorado foi compreender os efeitos da inoculação de Bacillus subtilis e Paenibacillus sp. em dois genótipos de palma forrageira, da espécie Nopalea cochenillifera Salm-Dick, sendo elas a ‘Baiana’ e a ‘Doce’ nas propriedades biológicas e microbioma da rizosfera nas condições edafoclimáticas do semiárido e sob déficit hídrico. No capítulo I e II foi conduzido um experimento a campo, em um delineamento inteiramente casualizado em parcelas subsubdivididas (2×3×2). A parcela principal foi o estágio de crescimento (90 e 270 dias) após o plantio da palma, enquanto a subparcela correspondeu à inoculação com BPCP (sem inoculação e inoculação com B. subtilis e Paenibacillus sp.). A sub-subparcela foi composta por dois genótipos de palma forrageira (‘Baiana’ e ‘Doce’). No capítulo I avaliou-se a biomassa microbiana de C, N e P, além da atividade enzimática, e concluiu-se que interação entre estágio&#13;
de crescimento, táxons de BPCP e genótipos de palma influenciam significativamente a&#13;
biomassa microbiana e a atividade enzimática na rizosfera, e que a combinação de genótipos específicos com B. subtilis pode aumentar a disponibilidade de fósforo. No capítulo II avaliou-se a diversidade, a composição e o enriquecimento das comunidades bacterianas na rizosfera, e concluiu-se que o microbioma da rizosfera da palma forrageira foi moldado principalmente pelo estágio de crescimento da planta, com grupos bacterianos-chave potencialmente associados à tolerância ao estresse. O capítulo III foi um experimento em casa de vegetação, em delineamento em blocos ao acaso, com arranjo fatorial 3 × 2. Os fatores adotados foram inoculação com BPCP (sem inoculação e inoculação com B. subtilis e Paenibacillus sp.) e duas&#13;
condições de disponibilidade hídrica (50% e 25% da capacidade de campo do solo). O genótipo de palma forrageira utilizado foi a ‘Miúda’ (equivalente ao genótipo ‘Doce’), e foram avaliados a estrutura, diversidade, abundância e redes de co-ocorrência da comunidade bacteriana da rizosfera. A partir dos resultados, concluiu-se que a disponibilidade de água alterou significativamente a estrutura da comunidade bacteriana, com Actinobacteriota dominando sob déficit hídrico e Proteobacteria sob condições bem irrigadas, além disso, B. subtilis promoveu redes microbianas cooperativas e pode potencialmente aumentar a resiliência à seca. ABSTRACT: In semiarid regions, drought-tolerant plants such as prickly pear cactus (family Cactaceae) areessential for sustaining food production for both humans and livestock. However, pro longed droughts intensified by climate change can compromise the growth of this plant. Some bacterial taxa, such as Bacillus and Paenibacillus, are known to colonize the rhizosphere and promote plant growth (PGPB), as well as mitigate the effects of water stress. Nevertheless, little is known about the effects of inoculation in cactus species on microbial biomass, enzymatic activity, and the rhizosphere microbiome under semiarid soil and climatic conditions. In this context, the aim of this doctoral thesis was to investigate the effects of inoculation with Bacillus subtilis and Paenibacillus sp. on two genotypes of prickly pear cactus (Nopalea cochenillifera Salm-Dick), ‘Baiana’ and ‘Doce’, and to evaluate their impacts on biological properties and the rhizosphere&#13;
microbiome under semiarid edaphoclimatic conditions and water deficit. In Chapters I and II, a field experiment was conducted using a completely randomized design in a split-split plot arrangement (2 × 3 × 2). The main plot factor was plant growth stage (90 and 270 days after planting), the subplot factor was PGPB inoculation (non-inoculated, and inoculated with B. subtilis or Paenibacillus sp.), and the sub-subplot factor consisted of the two cactus genotypes (‘Baiana’ and ‘Doce’). In Chapter I, microbial biomass C, N, and P, as well as enzymatic activities, were evaluated. The results showed that interactions among growth stage, PGPB taxa, and cactus genotypes significantly influenced microbial biomass and enzymatic activity in the&#13;
rhizosphere, and that combining specific genotypes with B. subtilis can enhance phosphorus availability. In Chapter II, the diversity, composition, and enrichment of rhizosphere bacterial communities were assessed. It was concluded that the rhizosphere microbiome of prickly pear cactus was shaped primarily by plant growth stage, with key bacterial groups potentially associated with stress tolerance. Chapter III describes a greenhouse experiment arranged in a randomized block design with a 3 × 2 factorial structure. The factors included PGPB inoculation (non-inoculated, inoculated with B. subtilis, or inoculated with Paenibacillus sp.) and two water availability conditions (50% and 25% of soil field capacity). The cactus genotype used was ‘Miúda’ (equivalent to the ‘Doce’ genotype), and the structure, diversity, abundance, and co-occurrence networks of the rhizosphere bacterial community were evaluated. The results&#13;
showed that water availability significantly altered bacterial community structure, with&#13;
Actinobacteriota dominating under water deficit and Proteobacteria under well-watered&#13;
conditions. Additionally, B. subtilis promoted cooperative microbial networks and may&#13;
potentially enhance drought resilience.

RESUMO: A caracterização citogenética e genômica é essencial para o estudo da diversidade vegetal e evolução dos recursos genéticos vegetais. Mimosa L. destaca-se por possuir uma rica biodiversidade endêmica do Cerrado brasileiro, contribuindo para a manutenção da sustentabilidade dos ecossistemas terrestres. O gênero Solanum L. possui destaque socioeconômico pela movimentação global de várias espécies importantes para a segurança alimentar mundial, em especial a batata (S. tuberosum L) e o tomate (S. lycopersicum L.). Apesar da relevância desses gêneros, suas espécies silvestres permanecem subexploradas, principalmente, sob a perspectiva citogenética e genômica. Neste trabalho, avaliou-se o cariótipo de diferentes populações de sete espécies de Mimosa e de doze espécies de Solanum mediante bandeamento por fluorocromos CMA/DAPI e estimativa do conteúdo de DNA por citometria de fluxo. Em Mimosa, eventos de polissomatia foram observados em seis espécies, exceto na espécie diploide M. verrucosa. O número de bandas de heterocromatina constitutiva (HC) CMA+ variou de quatro em M. caesalpiniifolia (células diploides) a 104 em M. candollei (células octaploides), sem aparente relação com a filogenia do gênero. O conteúdo de DNA variou de 1 C = 0,64 pg (M. sensitiva) a 1C = 1,55 pg (M. arenosa), sem relação com seu nível de ploidia (r = 0.1417). Os dados sugerem que a polissomatia é evento comum no gênero, possivelmente representando uma estratégia para adaptação dessas plantas ao ambiente, bem como seu crescimento e desenvolvimento. Nesse trabalho, realizou-se a primeira análise intra e interespecífica com relação ao padrão de HC e conteúdo de DNA para diferentes espécies de Mimosa, contribuindo para a compreensão da dinâmica da heterocromatina e organização genômica desse gênero cientificamente negligenciado. Em Solanum, reportamos novos dados de HC para S. andreanum, S. peruvianum, S. paludosum e valores de conteúdo de DNA 1C para S. corneliomullerii e S. andreanum, em um contexto filogenético. O número de blocos de HC variou de dois (S. viarum) a 42 (S. paludosum). Espécies do clado Potato geralmente exibiram maior número de blocos CH, enquanto em Lepstostemonum, geralmente exibiram poucos blocos, exceto S. paludosum. Contudo, não foi possível inferir a relação entre o padrão HC e os clados filogenéticos. O valor médio 1C variou de 1,0 pg em S. melongena (diploide) a 1,80 pg em S. laciniatum (octaploide), com correlação moderada (r = 0.69) para os níveis de ploidia das espécies analisadas. A caracterização citogenética de Mimosa e Solanum contribui, portanto, para a compreensão da sua estrutura cariotípica, distribuição do DNA repetitivo, estimativa do tamanho do genoma e dinâmica evolutiva dentro desses gêneros, fornecendo subsídios para estratégias de conservação e de programas de melhoramento genético de espécies de grande importância econômica, ambiental e social. ABSTRACT: Cytogenetics and genomics characterization is essential for understanding plant diversity and evolution of plant genetic resources. Mimosa L. stands out for its high biodiversity, most endemic to the Brazilian Cerrado, notably contributing to the sustainability of terrestrial ecosystems. Solanum L., in turn, has major socioeconomic importance due to the global importance of several species for world food security, particularly potato and tomato. Despite the relevance of both genera, their wild species remain largely underexplored, particularly from cytogenetic and genomic perspectives. In this study, we evaluated the karyotypes of different populations of seven Mimosa species and twelve Solanum species using CMA/DAPI fluorochrome banding and genome size estimation by flow cytometry. In Mimosa, polysomaty events were detected in six species, except for the diploid species M. verrucosa. The number of constitutive heterochromatin (CH) CMA⁺ bands ranged from four in M. caesalpiniifolia (diploid cells) to 104 in M. candollei (octaploid cells), with no apparent relationship to the phylogeny of the genus. Nuclear DNA content ranged from 1C = 0.64 pg (M. sensitiva) to 1C = 1.55 pg (M. arenosa), with no correlation with ploidy level (r = 0.1417). Our data indicates that polysomaty may be a common feature in the genus, possibly representing as an adaptive strategy related to growth, development and environmental conditions. This work provides the first intra- and interspecific analysis of CH patterns and DNA content across different Mimosa species, contributing to the understanding of heterochromatin dynamics and genome organization in this scientifically neglected genus. In Solanum, we report new CH data for S. andreanum, S. peruvianum, and S. paludosum, as well as novel 1C DNA content estimates for S. corneliomullerii and S. andreanum, in a phylogenetic context. The number of CH blocks ranged from two (S. viarum) to 42 (S. paludosum). Species of the Potato clade generally exhibited a higher number of CH blocks, whereas members of Leptostemonum clade exhibited fewer blocks, except for S. paludosum. However, we could not infer a clear relationship between the CH pattern and the phylogenetic clades. Mean of 1C value ranged from 1.0 pg in S. melongena (diploid) to 1.80 pg in S. laciniatum (octaploid), revealing a moderate correlation with the ploidy levels (r = 0.69). The cytogenetic characterization of Mimosa and Solanum enhances our understanding of karyotype structure, repetitive DNA distribution, genome size estimation and evolutionary dynamics within these genera, providing valuable information for conservation strategies and breeding programs of species with significant ecological, economic, and social importance.

RESUMO: Tetranychus gloveri (Acari: Tetranychidae) emergiu como uma praga  de relevância crescente em sistemas agrícolas e ornamentais, especialmente em regiões tropicais. O objetivo deste trabalho foi relatar  a ocorrência de T. gloveri em Adenium spp., caracterizar injúrias foliares e adequação de espécies do gênero como hospedeiras via crescimento populacional, bem como avaliar um método sustentável para seu manejo, através da investigação do potencial acaricida de óleos essenciais. Inicialmente, foi registrada a ocorrência de T. gloveri em espécies do gênero Adenium e avaliada sua multiplicação em Adenium obesum, A. arabicum e A. somalense, através da quantificação de injúrias foliares e estimativa da taxa instantânea de crescimento populacional (ri). Em seguida, foram utilizados os óleos essenciais de Zingiber officinale, Rosmarinus officinalis e Foeniculum vulgare, sendo avaliados quanto à toxicidade através de contato residual, repelência, efeito ovicida e impactos sobre ri. Este estudo revela a primeira ocorrência de de T. gloveri em Adenium spp no Brasil., com injúrias variando de 61% a 80% da área foliar e nota máxima de dano 5, além de valores positivos de ri em todas as espécies, indicando potencial reprodutivo e aptidão dessas plantas como hospedeiras. A análise química revelou zingibereno como majoritário em Z. officinale, 1,8-cineol e α-pineno em R. officinalis e trans-anetol em F. vulgare. Nos bioensaios, Z. officinale apresentou a maior toxicidade (CL50 = 12,97 µL/mL), seguido por R. officinalis (14,94 µL/mL) e F. vulgare (24,91 µL/mL). De forma integrada, Z. officinale destacou-se apresentando  elevada toxicidade e redução da ri, R. officinalis apresentou ação intermediária, enquanto F. vulgare exerceu papel relevante na repelência e na redução da oviposição e ri. Conclui-se que Adenium spp. são hospedeiras favoráveis a T. gloveri e que os óleos essenciais apresentam potencial para o manejo desse ácaro. ABSTRACT: Tetranychus gloveri (Acari: Tetranychidae) has emerged as a mite of increasing relevance in agricultural and ornamental systems, particularly in tropical regions. The objective of this study was to record the occurrence of T. gloveri on ornamental plants, characterize leaf injuries, and assess the suitability of Adenium spp. as hosts based on population growth, as well as to evaluate a sustainable management approach through the investigation of the acaricidal potential of essential oils. Initially, the occurrence of the mite was recorded in species of the genus Adenium, and its population increase was evaluated on Adenium obesum, A. arabicum, and A. somalense by quantifying leaf injuries and estimating the instantaneous rate of population growth (ri). Subsequently, essential oils from Zingiber officinale, Rosmarinus officinalis, and Foeniculum vulgare were used, with their chemical composition determined by gas chromatography–mass spectrometry, and evaluated for residual contact toxicity, repellency, ovicidal effect, and impacts on ri. The results demonstrated, for the first time in Brazil, the occurrence of T. gloveri on Adenium spp., with leaf injuries ranging from 61% to 80% of the leaf area and a maximum damage score of 5, as well as positive ri values for all species, indicating reproductive potential and host suitability. Chemical analysis revealed zingiberene as the major compound in Z. officinale, 1,8-cineole and α-pinene in R. officinalis, and transanethole in F. vulgare. In bioassays, Z. officinale showed the highest toxicity (LC₅₀ = 12.97 µL/mL), followed by R. officinalis (14.94 µL/mL) and F. vulgare (24.91 µL/mL). Overall, Z. officinale stood out for its high toxicity and reduction in ri, R. officinalis exhibited intermediate activity, whereas F. vulgare played a relevant role in repellency and in reducing oviposition and ri. It is concluded that Adenium spp. are suitable hosts for T. gloveri and that essential oils show potential for the management of this mite.

RESUMO: O feijão - caupi é cultivado em regiões tropicais e subtropicais no mundo devido a sua&#13;
importância sócioeconômica. No seu armazenamento, o caruncho causa danos que&#13;
podem chegar a 100% em pouco tempo, inviabilizando para consumo, semeio e&#13;
comercialização. O controle do caruncho é realizado com produto químico, fosfina,&#13;
altamente tóxico. Como alternativa ao uso de químico, tem-se buscado por métodos&#13;
mais sustentáveis como a resistência de plantas. O objetivo deste trabalho foi&#13;
selecionar e identificar a causa da resistência de genótipos de feijão- caupi ao&#13;
caruncho Calosobruchus maculatus. 60 genótipos de feijão-caupi foram utilizados em&#13;
teste sem chance de escolha, avaliando-se total de ovos, ovos viáveis, ovos inviáveis,&#13;
viabilidade de ovos (%), insetos emergidos, percentual de insetos emergidos, período&#13;
de ovo a adulto, massa consumida (g) e emergência precoce de larvas. Com base&#13;
nesses resultados foram selecionados 06 genótipos com os quais foram realizados&#13;
experimentos sem chance de escolha: com sementes naturais onde foram avaliados&#13;
total de ovos por sementes, sobrevivência larval(%), massa larval (mg), comprimento&#13;
larval (mm) e feito registro fotográfico; no experimento com sementes artificiais a partir&#13;
das farinhas dos cotilédones dos genótipos de feijão-caupi foram avaliados&#13;
sobrevivência larval (%), massa larval (mg), comprimento larval (mm), total de adultos&#13;
emergidos (%) e emergência de adultos (dias) e registro fotográfico das larvas.&#13;
Posteriormente, foram isoladas as proteínas de ligação à quitina destes genótipos&#13;
para avaliar a toxicidade sobre C. maculatus em experimento sem chance de escolha&#13;
com sementes artificiais contendo a fração de proteínas que se ligam à quitina a 1%&#13;
e a 5%, avaliando-se sobrevivência larval (%), massa larval (mg), comprimento larval&#13;
&#13;
e feito registro fotográfico das larvas. Os genótipos IT81D-1045, IT86D-716-1, IT85F-&#13;
2687, Monteiro, MNC11-1020E-16 e MNC05828B-3-15 apresentaram antibiose a C.&#13;
&#13;
maculatus, pois reduziram a emergência de insetos e viabilizaram uma redução de&#13;
emergência acima de 40%. Em experimento com sementes naturais os genótipos&#13;
Monteiro e IT85F-2687 reduziram a sobrevivência larval, os genótipos IT85F-2687 e&#13;
&#13;
MNC11-1020E-16 reduziram a massa larval e os genótipos MNC11-1020-16 e IT85F-&#13;
2687 reduziram o comprimento larval. Em sementes artificiais com farinha de&#13;
&#13;
cotilédones dos genótipos, o genótipo IT85F-2687 apresentou a maior sobrevivência&#13;
larval (100%), o maior comprimento larval foi encontrado no genótipo Monteiro,&#13;
todavia, a emergência de insetos foi reduzida pelo genótipo IT85F-2687 e os maiores&#13;
atrasos na emergência de insetos ocorreram nos genótipos IT86D716-1 e IT85F-2687.&#13;
No experimento com sementes artificiais a fração proteica de 5% foi toxica às larvas&#13;
de C. maculatus, causando 100% de mortalidade, enquanto a fração proteica de 1%&#13;
dos genótipos MNC05-828B-3-15; IT86D-716-1, IT85F-2687 e MNC11- 1020E-16&#13;
reduziu a massa e comprimento larval. Os genótipos selecionados IT85F-2687,&#13;
IT86D-716-1, MNC11-1020E-16 e MNC05828-3-15 apresentam resistência a C.&#13;
maculatus, que está relacionada com proteínas que se ligam à quitina e são tóxicas&#13;
ao referido inseto. ABSTRACT: Cowpea bean is cultivated in tropical and subtropical regions of the world due to its&#13;
socioeconomic importance. During its storage, the weevil causes damage that can&#13;
reach 100% in a short period of time, making it unviable for consumption, sowing and&#13;
marketing. Weevil control is made with phosphine, a highly toxic Chemical. As an&#13;
alternative to the use of chemicals, more sustainable methods have been sought, such&#13;
as plant resistance. The objective of this work was to select and identify the cause of&#13;
resistance of cowpea bean genotypes to the weevil Calosobruchus maculatus. 60&#13;
cowpea genotypes were used in a no-choice test, evaluating total eggs, viable eggs,&#13;
nonviable eggs, egg viability (%), emerged insects, percentage of emerged insects,&#13;
period from egg to adult, mass consumed (g) and premature emergence of larvae.&#13;
Based on these results, 6 genotypes were selected in which no-choice experiments&#13;
were made: the total number of eggs per seed, larval survival (%), larval mass (mg),&#13;
and larval length (mm) of natural seeds were evaluated, and photographic records&#13;
were made; in the experiment with artificial seeds from cotyledon flours of cowpea&#13;
genotypes, larval survival (%), larval mass (mg), larval length (mm), total number of&#13;
emerged adults (%) and adult emergence (days), were evaluated and photographic&#13;
records of the larvae were made. Subsequently, the chitin-binding proteins of these&#13;
genotypes were isolated to evaluate their toxicity on C. maculatus in a no-choice&#13;
experiment with artificial seeds containing the fraction of proteins that bind to chitin at&#13;
1% and 5%, evaluating larval survival (%), larval mass (mg), larval length and&#13;
&#13;
photographic records of the larvae. The genotypes IT81D-1045, IT86D-716-1, IT85F-&#13;
2687, Monteiro, MNC11-1020E-16 and MNC05828B-3-15 showed antibiosis to C.&#13;
&#13;
maculatus, as they reduced insect emergence and enabled an emergence reduction&#13;
&#13;
above 40%. In an experiment with natural seeds, the genotypes Monteiro and IT85F-&#13;
2687 reduced larval survival, the genotypes IT85F-2687 and MNC11-1020E-16&#13;
&#13;
reduced larval mass, and the genotypes MNC11-1020-16 and IT85F-2687 reduced&#13;
larval length. In artificial seeds with cotyledon flour of the genotypes, the IT85F-2687&#13;
genotype showed the highest larval survival (100%), the greatest larval length was&#13;
found in the Monteiro genotype; however, insect emergence was reduced by the&#13;
IT85F-2687 genotype, and the greatest delays in insect emergence occurred in the&#13;
IT86D716-1 and IT85F-2687 genotypes. In the experiment with artificial seeds, the 5%&#13;
protein fraction was toxic to C. maculatus larvae, causing 100% mortality, while the 1%&#13;
protein fraction of the genotypes MNC05-828B-3-15; IT86D-716-1, IT85F-2687 and&#13;
&#13;
MNC11-1020E-16 reduced larval mass and length. The selected genotypes IT85F-&#13;
2687, IT86D-716-1, MNC11-1020E-16 and MNC05828-3-15 present resistance to C.&#13;
&#13;
maculatus, which is related to proteins that bind to chitin and are toxic to the insect.
