<title>AVALIAÇÃO DA TOXICIDADE DO ÓXIDO DE ROSA EM MODELOS IN SILICO, IN VITRO E IN VIVO.</title>

<summary type="text">AVALIAÇÃO DA TOXICIDADE DO ÓXIDO DE ROSA EM MODELOS IN SILICO, IN VITRO E IN VIVO.

RESUMO: O óxido de rosa é um monoterpeno que possui atividade anti-inflamatória e antioxidante. No entanto, não existem testes de toxicidade a cerca desse composto. Com isso, o estudo teve objetivo avaliar a toxicidade do Óxido de Rosa em modelos in silico, in vivo e in vitro. Para avaliação in silico, foram utilizadas as plataformas ADMETLab 3.0 e a SwissADME, nas quais o smile da substância foi adicionado e a partir disso, dados físico-químicos e de toxicidade foram coletados. Para avaliação in&#13;
vivo da toxicidade em larvas de Zophobas morio, a análise foi feita com diluição da substância para obtenção das doses estabelecidas que foram administradas na hemocele do tenébrio. Na avaliação in vitro, no teste de hemólise foram utilizados eritrócitos de Canis lupus familiaris, com análise em triplicata e quantificada a presença de hemoglobina. Ademais, foi realizado bioensaio de letalidade de Artemia salina, para avaliar a toxicidade do óxido de rosa, conforme metodologia de Batista et al. No que diz respeito aos resultados, conclui-se que o Óxido de Rosa de acordo com o estudo in silico, não apresenta mutagenicidade, nem carcinogenicidade, classificando-se na categoria II no que diz respeito à Toxicidade Oral Aguda, apresentando média toxicidade e um bom perfil de segurança. No que se refere ao estudo in vivo realizado com larvas de Zophobas morio, infere-se que o Óxido de Rosa apresentou baixa toxicidade no teor de melanização, bem como na taxa de sobrevivência. Outrossim, o composto analisado não apresentou atividade hemolítica considerável no teste in vitro. No que diz respeito ao teste de Artemia Salina, o óxido de rosa não apresentou efeitos letais significativos, indicando ausência de toxicidade aguda. Assim sendo, conclui-se que o Óxido de Rosa puro é um possivel candidato a fármaco promissor no futuro, pois possui baixa toxicidade e um bom perfil de segurança. ABSTRACT: Rose oxide is a monoterpene with anti-inflammatory and antioxidant activity. However, there are no&#13;
toxicity tests for this compound. Therefore, the study aimed to evaluate the toxicity of rose oxide in in silico, in vivo, and in vitro models. For the in silico evaluation, the ADMETLab 3.0 and SwissADME platforms were used, to which the substance's smile was added. From this, physicochemical and toxicity data were collected. For the in vivo toxicity evaluation in Zophobas morio larvae, the analysis was performed by diluting the substance to obtain the established doses that were administered into the mealworm hemocoel. In the in vitro hemolysis test, Canis lupus familiaris erythrocytes were used,&#13;
analyzed in triplicate, and the presence of hemoglobin was quantified. Furthermore, a lethality bioassay was performed on Artemia salina to evaluate the toxicity of rose oxide, according to the methodology of Batista et al. The results concluded that rose oxide, according to the in silico study, is neither mutagenic nor carcinogenic, classifying it as Category II for Acute Oral Toxicity, exhibiting moderate toxicity and a good safety profile. Regarding the in vivo study conducted with Zophobas morio larvae, it was inferred that rose oxide presented low toxicity in terms of melanization level and survival rate.&#13;
Furthermore, the compound analyzed did not exhibit significant hemolytic activity in the in vitro test. Regarding the Artemia Salina test, rose oxide did not exhibit significant lethal effects, indicating the absence of acute toxicity. Therefore, it is concluded that pure rose oxide is a potential promising drug candidate in the future, as it has low toxicity and a good safety profile.

RESUMO: Estima-se que 4,05% da população mundial (301 milhões) possua um transtorno de ansiedade. A ansiedade torna-se patológica quando os comportamentos de evitação e o medo sustentam- se e são perturbadores. A depressão, por sua vez, atinge aproximadamente 3,8% da população. Devido aos efeitos adversos dos ansiolíticos e antidepressivos convencionais, necessita-se de alternativas mais eficazes e com menos efeitos adversos na farmacoterapia desses transtornos.&#13;
Uma alternativa estaria no uso de produtos naturais, como os alcalóides indólicos, conhecidos como psicodélicos clássicos. A Mimosa tenuiflora (Wild) Poir. (Jurema Preta), planta típica de áreas semiáridas do Brasil, possui alcalóides psicodélicos, como N, N-dimetiltriptamina (DMT), com atividades farmacológicas provavelmente mediadas por receptores serotoninérgicos. Este trabalho teve como objetivo avaliar o efeito ansiolítico e antidepressivo da fração alcaloídica II (FA II) proveniente do extrato da casca da raiz da M. tenuiflora em estudos in sílico e in vivo com camundongos C57BL/6. Nos estudos in sílico, foi utilizada a plataforma PreADMET. A Bufotenina e o DMT atravessam livremente a BHE, e a Yuremamina atravessa de forma moderada. Os três compostos da FA II apresentaram média permeabilidade nas células Caco-2. Em relação à absorção intestinal, todas demonstraram níveis elevados. Os valores de ligação às proteínas plasmáticas indicaram que a Bufotenina e o DMT possuem baixa&#13;
ligação, e a Yuremamina apresenta ligação elevada. Nenhuma das substâncias foi capaz de inibir a P-gp. A Bufotenina e o DMT são substratos fracos da CYP3A4 e não inibem essa enzima, no entanto, a Yuremamina, demonstrou capacidade de inibição. Em relação ao potencial carcinogênico, os três compostos apresentaram resultados negativos em camundongos. Na inibição do gene hERG, a Bufotenina e o DMT demonstraram risco médio, e a Yuremamina apresentou risco alto. Todas os compostos apresentaram resultados negativos para mutagenicidade nas cepas TA1535 +S9, TA100 +S9 e TA100 -S9, exceto a Yuremamina, que apresentou resultado positivo para TA 1535 -S9. Foi realizado o docking molecular para prever a interação entre os constituintes da FA II e seus alvos biológicos, demonstrando que os compostos da FA II foram capazes de interagir com os sítios ativos dos alvos serotoninérgicos. Nos testes comportamentais, foram realizados o Teste do Labirinto em Cruz Elevado (TLCE), Teste da Caixa Claro-Escuro (TCCE), Teste do Campo Aberto (TCA), Teste de suspensão pela cauda (TSC) e RotaRod, com as doses 3, 6 e 9 mg/kg da FA II. Os resultados foram apresentados como média ± erro padrão da média (E.P.M.) e analisados por ANOVA, seguido pelo teste post hoc de Dunnett, utilizando o software GraphPad (versão 8.0). Considerou-se significância estatística para valores de p &lt; 0,05. No TCLE, a FA II na dose de 9 mg/kg apresentou aumento no número de entradas nos braços abertos, e as doses de 3, 6 e 9 mg/kg aumentaram o tempo de permanência dos camundongos nos braços abertos. No TCCE, a FA II reduziu o tempo de latência, indicando efeito ansiolítico. No TCA, a melhor resposta foi observada nas três doses testadas, que reduziram o grooming. No TSC, a FA II reduziu o tempo&#13;
de imobilidade dos animais em todas as doses. No RotaRod, a dose de 3 mg/kg aumentou o número de quedas dos animais, e as demais doses não alteraram significativamente o tempo de permanência no aparelho. Os testes in sílico demonstraram resultados promissores nos parâmetros estudados. Os testes comportamentais demonstraram que a FA II possui um efeito ansiolítico e antidepressivo. ABSTRACT: It is estimated that 4.05% of the world's population (301 million) has an anxiety disorder. Anxiety becomes pathological when avoidance behaviors and fear are sustained and disruptive. Depression, in turn, affects approximately 3.8% of the population. Due to the adverse effects of conventional anxiolytics and antidepressants, more effective alternatives with fewer adverse&#13;
effects are needed in the pharmacotherapy of these disorders. One alternative would be the use of natural products, such as indole alkaloids, known as classic psychedelics. Mimosa tenuiflora (Wild) Poir. (Jurema Preta), a plant typical of semiarid areas of Brazil, has psychedelic alkaloids, such as N,N-dimethyltryptamine (DMT), with pharmacological activities probably mediated by serotonergic receptors. This study aimed to evaluate the anxiolytic and antidepressant effects of the alkaloid fraction II (FA II) from the root bark extract of M. tenuiflora in in silico and in vivo studies with C57BL/6 mice. In the in silico studies, the PreADMET platform was used. Bufotenine and DMT freely cross the BBB, and Yuremamine crosses moderately. The three FA II compounds showed medium permeability in Caco-2 cells. Regarding intestinal absorption, all showed high levels. Plasma protein binding values indicated that Bufotenine and DMT have low binding, and Yuremamine has high binding. None of the substances was able to inhibit P-gp. Bufotenine and DMT are weak substrates of CYP3A4 and do not inhibit this enzyme; however, Yuremamine demonstrated inhibition capacity. Regarding carcinogenic potential, the three compounds showed negative results in mice. In the inhibition&#13;
of the hERG gene, Bufotenine and DMT demonstrated medium risk, and Yuremamine&#13;
presented high risk. All compounds presented negative results for mutagenicity in the strains TA1535 +S9, TA100 +S9 and TA100 -S9, except Yuremamine, which presented positive results for TA 1535 -S9. Molecular docking was performed to predict the interaction between the constituents of FA II and their biological targets, demonstrating that the compounds of FA II were able to interact with the active sites of the serotonergic targets. In the behavioral tests, the Elevated Plus Maze Test (ELMT), Light-Dark Box Test (LDCT), Open Field Test (OFT), Tail Suspension Test (TSC) and RotaRod were performed, with doses of 3, 6 and 9 mg/kg of FA II. The results were presented as mean ± standard error of the mean (S.E.M.) and analyzed by ANOVA, followed by Dunnett's post hoc test, using GraphPad software (version 8.0). Statistical significance was considered for values of p &lt; 0.05. In the TCLE, FA II at a dose of 9 mg/kg increased the number of entries into the open arms, and the doses of 3, 6 and 9 mg/kg increased the time the mice remained in the open arms. In the TCCE, FA II reduced the latency time, indicating an anxiolytic effect. In the ACT, the best response was observed in the three doses tested, which reduced grooming. In the TSC, FA II reduced the immobility time of the&#13;
animals in all doses. In the RotaRod, the dose of 3 mg/kg increased the number of falls of the animals, and the other doses did not significantly alter the time spent in the device. The in silico tests showed promising results in the parameters studied. Behavioral tests have demonstrated that FA II has an anxiolytic and antidepressant effect.

RESUMO: As plantas desde muito tempo são utilizadas na medicina popular, substâncias purificadas e isoladas a partir da matéria-prima vegetal, com estrutura química definida e atividade farmacológica, são denominadas fitofármacos e podem ser usados como ativos em medicamentos com propriedade profilática, paliativa ou&#13;
curativa. Têm-se aumentado o interesse em explorar propriedades bioquímicas&#13;
dos metabólicos secundários (MS), os flavonoides são exemplos desses&#13;
metabólitos e possuem inúmeras propriedades como antioxidantes,&#13;
antiinflamatórias, anticancerígenas, antivirais e analgésicas. Além de serem&#13;
responsáveis pela ação farmacológica da planta, alguns desses MS, também&#13;
podem ser tóxicos, assim testes de toxicidade são de extrema importância e&#13;
nesse contexto o estudo teve objetivo de avaliar a toxicidade da esterubina em&#13;
modelos in sílico, in vitro e in vivo. Para avaliação in sílico foi utilizada a&#13;
plataforma ADMETLab 3.0, onde o smile da substância foi adicionado e a partir&#13;
disso dados físico-químicos e de toxicidade foram coletados. Para avaliação in&#13;
vitro da toxicidade foi realizado o teste de hemólise, onde foram utilizados&#13;
eritrócitos de Canis lupus familiaris, com análise em triplicata as amostras foram&#13;
centrifugadas a 3000 rpm por 5 minutos e quantificada a presença de&#13;
hemoglobina, na análise in vitro da toxicidade pelo método MTT foram utilizadas&#13;
linhagens celulares L929 (fibroblastos de camundongos) e B16F10 (melanoma&#13;
murino) e avaliados quanto a viabilidade celular. Para realização dos testes in&#13;
vivo foram utilizadas larvas de Zophobas morio a análise foi feita em duplicata&#13;
com diluição da substância para obtenção das doses de 3, 30 e 300 mg/kg, que&#13;
são doses já preconizadas para o teste de toxicidade, e foram administradas na&#13;
hemocele do tenébrio sendo verificadas a melanização e sobrevivência. A&#13;
previsão de toxicidade pelo ADMETlab 3.0 trouxe resultados promissores,&#13;
mostrando a molécula de esterubina com valores favoráveis de propriedades&#13;
como genotoxicidade, mutagenicidade no teste AMES, irritação ocular, e valores&#13;
com toxicidade média que não impedem seu uso, mas merecem atenção&#13;
especial como carcinogenicidade. No teste de hemólise os resultados foram&#13;
positivos apresentando porcentagem menor que 10%. Na avaliação de&#13;
citotoxicidade pelo método MTT, a molécula teve eficácia em reduzir a&#13;
viabilidade quando comparadas ao controle negativo assim como também o&#13;
valor de IC50 de 15,07 μg/mL (12,55 a 18,10 μg/mL) em células B16F10,&#13;
mostrando também um bom potencial em reduzir a viabilidade nessa linhagem&#13;
celular. No teste in vivo com larvas de Zophobas morio apresentou boa taxa de&#13;
sobrevivência em doses maiores de 30 e 300 mg/kg, demonstrando segurança.&#13;
Com base nos resultados a esterubina se mostra uma molécula promissora com&#13;
bom perfil de segurança demonstrado nos testes in sílico, in vitro e in vivo se&#13;
mostrando segura para futuros testes in vivo que possam vir a serem realizados&#13;
para evidenciar suas possíveis atividades farmacológicas. ABSTRACT: Plants have long been used in folk medicine. Purified substances isolated from&#13;
plant raw materials, with a defined chemical structure and pharmacological&#13;
activity, are called phytopharmaceuticals and can be used as active ingredients&#13;
in medicines with prophylactic, palliative or curative properties. There has been&#13;
increasing interest in exploring the biochemical properties of secondary&#13;
metabolites (SM). Flavonoids are examples of these metabolites and have&#13;
numerous properties such as antioxidants, anti-inflammatory, anticancer, antiviral&#13;
and analgesics. In addition to being responsible for the pharmacological action of&#13;
the plant, some of these SM can also be toxic, so toxicity tests are extremely&#13;
important and in this context the study aimed to evaluate the toxicity of sterubin&#13;
in in silico, in vitro and in vivo models. For in silico evaluation, the ADMETLab 3.0&#13;
platform was used, where the substance's smile was added and from this,&#13;
physicochemical and toxicity data were collected. For in vitro toxicity evaluation,&#13;
the hemolysis test was performed, where Canis lupus familiaris erythrocytes were&#13;
used, with analysis in triplicate. The samples were centrifuged at 3000 rpm for 5&#13;
minutes and the presence of hemoglobin was quantified. In the in vitro toxicity&#13;
analysis by the MTT method, L929 (mouse fibroblasts) and B16F10 (murine&#13;
melanoma) cell lines were used and evaluated for cell viability. To perform the in&#13;
vivo tests, Zophobas morio larvae were used. The analysis was performed in&#13;
duplicate with dilution of the substance to obtain doses of 3, 30 and 300 mg/kg,&#13;
which are doses already recommended for the toxicity test, and were&#13;
administered in the hemocoel of the tenebrio, verifying melanization and survival.&#13;
The toxicity prediction by ADMETlab 3.0 brought promising results, showing the&#13;
sterubin molecule with favorable values of properties such as genotoxicity,&#13;
mutagenicity in the AMES test, eye irritation, and values with medium toxicity that&#13;
do not prevent its use, but deserve special attention as carcinogenicity. In the&#13;
hemolysis test, the results were positive, with a percentage lower than 10%. In&#13;
the cytotoxicity evaluation by the MTT method, the molecule was effective in&#13;
reducing viability when compared to the negative control, as well as the IC50 value&#13;
of 15.07 μg/mL (12.55 to 18.10 μg/mL) in B16F10 cells, also showing good&#13;
potential in reducing viability in this cell line. In the in vivo test with Zophobas&#13;
morio larvae, it showed a good survival rate at doses higher than 30 and 300&#13;
mg/kg, demonstrating safety. Based on the results, sterubin is a promising&#13;
molecule with a good safety profile demonstrated in in silico, in vitro and in vivo&#13;
tests, proving safe for future in vivo tests that may be performed to demonstrate&#13;
its possible pharmacological activities.

<title>PERFIL TOXICOGENÉTICO E ANTIPROLIFERATIVO DA CRISINA LIVRE EM TESTES NÃO CLÍNICOS</title>

<summary type="text">PERFIL TOXICOGENÉTICO E ANTIPROLIFERATIVO DA CRISINA LIVRE EM TESTES NÃO CLÍNICOS

RESUMO: O câncer é uma doença responsável por inúmeras mortes ao ano e um problema global. Desta forma, é importante investigar novos compostos terapêuticos mais seletivos, que possam ajudar a minimizar esse cenário. As investigações toxicológicas de substâncias são essenciais no processo de estudo de fármacos, pois permitem direcionar outras investigações que possam contribuir para a descoberta de&#13;
substâncias com atividade antitumoral. Dentro desse contexto, a crisina é um&#13;
flavonoide de origem natural com propriedades biológicas de importância terapêutica&#13;
como atividade anti-inflamatória, antioxidante, antigenotóxica, apoptótica, entre&#13;
outros. Visando contribuir para as possibilidades de aplicação da crisina dentro do&#13;
contexto oncológico, o presente estudo teve o objetivo de avaliar sua toxicidade em&#13;
teste de A. salina, seus possíveis efeitos genotóxicos em células de A. cepa, efeitos&#13;
antiproliferativos em células tumorais e não tumorais, bem como prever seu perfil de&#13;
semelhança à fármacos e toxicidade em análise in silico. A toxicidade da crisina&#13;
avaliada por Artemia salina nos períodos de 24h e 48h tiveram a CL50 determinada&#13;
em 657,0 (528,8 – 866,2 μM) e 258,9 (193,6 – 367,7μM) respectivamente. No ensaio&#13;
de Allium cepa, a crisina apresentou efeito citotóxico nas duas maiores concentrações&#13;
entre as avaliadas (250, 125 e 62,5 μM), entretanto, não foi identificado efeitos&#13;
genotóxicos. O efeito antiproliferativo com base no ensaio de MTT frente as linhagens&#13;
MCF-7 e MCF-10 apresentou valores de IC50 de 53,4 (23,7 – 133,5 μM) e 120,8 (39,53&#13;
– 785,1 μM) respectivamente. Não foi possível determinar o valor de IC50 nas&#13;
linhagens B16F10 e U87 sendo estimando como superior as maiores concentrações&#13;
utilizadas (&gt;125 μM e &gt;200 μM respectivamente). Na linhagem MCF-7, a combinação&#13;
da crisina com os quimioterápicos demonstrou um aumento no efeito antiproliferativo&#13;
através da diminuição dos valores de IC50 sendo para cisplatina de 9,4 (8,55 -14,46&#13;
μM) para 5,3 (4,17 - 6,3 μM) e para doxorrubicina de 2,5 (1,91 – 3,36 μM) para 0,3&#13;
(0,25 - 0,40 μM). As previsões in silico demonstraram que a crisina não viola nenhuma&#13;
das regras pré-estabelecidas para compostos candidatos à fármacos e apresentou&#13;
toxicidade relativa baixa, além disso, a ancoragem molecular indicou a interação da&#13;
crisina com os respectivos alvos: CDK1 = -8,1; CDK2 = -10,2; CDK6 = -8,5; EGFR = -&#13;
7,8; VEGFR2 = -9,0; ER-α = -8,2 e CYP19A1 = -8,3 (kcal/moL). Diante dos resultados,&#13;
conclui-se que a crisina apresenta efeitos citotóxicos que podem explicar sua ação&#13;
antiproliferativa em células tumorais. Além disso as previsões in silico ainda&#13;
demonstram que ela possui, em uma análise inicial, características físico-químicas&#13;
fundamentais para fármacos com biodisponibilidade oral compatível e potencial de&#13;
inibição para alvos importantes no câncer de mama. ABSTRACT: Cancer is responsible for numerous deaths annually and represents a global health&#13;
problem. Therefore, it is crucial to investigate novel and, more selective therapeutic&#13;
compounds that may help mitigate this scenario. Toxicological investigations of&#13;
substances are essential in drug research, as they guide further studies that may&#13;
contribute to discovery of compounds with antitumor activity. Within this context,&#13;
chrysin is a naturally occurring flavonoid with biologically significant therapeutic&#13;
properties, such as anti-inflammatory, antioxidant, antigenotoxic, and apoptotic&#13;
activities, among others. To explore the potential applications of chrysin in oncology,&#13;
this study aimed to evaluate its toxicity using the A. salina assay, its possible genotoxic&#13;
effects in A. cepa cells, its antiproliferative effects in tumor and non-tumor cells, as well&#13;
as predict its drug-likeness profile and toxicity through in silico analysis. The toxicity of&#13;
chrysin, assessed in Artemia salina at 24h and 48h, had LC50 values of 657.0 (528.8&#13;
– 866.2 μM) and 258.9 (193.6 – 367.7 μM), respectively. In the Allium cepa assay,&#13;
chrysin presented cytotoxic effects at the two highest tested concentrations (250, 125,&#13;
and 62.5 μM); however, no genotoxic effects were identified. The antiproliferative&#13;
effect, based on the MTT assay in the MCF-7 and MCF-10 cell lines, showed IC50&#13;
values of 53.4 (23.7 – 133.5 μM) and 120.8 (39.53 – 785.1 μM), respectively. It was&#13;
not possible to determine the IC50 values for the B16F10 and U87 cell lines, as they&#13;
were estimated to be above the highest concentrations used (&gt;125 μM and &gt;200 μM,&#13;
respectively). In the MCF-7 cell line, the combination of chrysin with chemotherapeutic&#13;
agents enhanced the antiproliferative effect by reducing IC50 values: for cisplatin, from&#13;
9.4 (8.55 – 14.46 μM) to 5.3 (4.17 – 6.3 μM), and for doxorubicin, from 2.5 (1.91 – 3.36&#13;
μM) to 0.3 (0.25 – 0.40 μM). In silico predictions indicated that chrysin does not violate&#13;
any pre-established rules for drug-like compounds and exhibited relatively low toxicity.&#13;
Additionally, molecular docking analysis showed interactions between chrysin and the&#13;
following targets: CDK1 = -8.1; CDK2 = -10.2; CDK6 = -8.5; EGFR = -7.8; VEGFR2 =&#13;
-9.0; ER-α = -8.2; and CYP19A1 = -8.3 (kcal/mol). Based on these results, it is&#13;
concluded that chrysin exhibits cytotoxic effects that may explain its antiproliferative&#13;
action in tumor cells. Furthermore, in silico predictions suggest that chrysin possesses&#13;
key physicochemical properties for oral bioavailability and potential inhibition of&#13;
important targets in breast cancer.
