Abstract:
RESUMO:Introdução: Atrofia vaginal e o câncer mamário hormônio sensível são condições
comuns em mulheres pós - menopáusicas e o tamoxifeno, por sua vez, é a terapia
endócrina padrão para esta doença, todavia apesar de mostrar efeito trófico vaginal
pode provocar reações endometriais graves, daí a busca de endocrinoterapia
alternativa. Objetivo: Avaliar a expressão do antígeno Ki-67 no epitélio vaginal de
ratas em estro permanente tratadas com anastrazol. Materiais e métodos:
Selecionadas 42 ratas induzidas ao estado de estro permanente e dividas em 2
grupos: grupo I (controle, n=21) o qual foi administrado 0,1 ml/animal/dia de
propilenoglicol e grupo II (experimental n=21) receberam 0,5 mg/Kg de anastrazol
diluído em 0,1ml de propilenoglicol. Avaliação imunoistoquímica do marcador Ki-67
foi realizada com análise microscópica as células do epitélio vaginal, à análise
estatística, foi realizada utilizando o teste não paramétrico Mann Whitney.
Resultados: A microscopia de luz mostrou uma maior concentração de células com
núcleo corados em marrom com anti-Ki-67 no grupo controle do que no grupo
experimental. A porcentagem média de núcleos corados com Ki-67 por 500 células
no epitélio vaginal foi de 68,64 ± 2,64 e 30,46 ± 2,00 [média +/- erro padrão da
média (SEM)] nos grupos controle e experimental, respectivamente (p <0,003).
Conclusões: O presente estudo mostrou que o anastrazol, na dose e duração do
tratamento utilizado, induziu uma diminuição significativa na proliferação celular da
mucosa vaginal de ratas no estro permanente, avaliado pela expressão da proteína
Ki-67.
ABSTRACT:Introduction: Vaginal atrophy and breast cancer sensitive hormone are common
conditions in postmenopausal women and tamoxifen, in turn, is the standard
endocrine therapy for this disease, although despite showing vaginal trophic effect
can cause severe endometrial reactions, hence the search for alternative endocrine
therapy. Objective: To evaluate Ki-67 antigen expression in the vaginal epithelium of
rats treated with anastrozole in permanent estrus. Materials and methods: 42 rats
were induced to the state of permanent estrus and divided into 2 groups: group I
(control, n = 21), which was administered 0.1 ml/animal/day of propylene glycol and
group II (experimental n = 21) will receive 0.5 mg/kg or 0.125 mg/animal/day of
anastrozole diluted in 0.1 ml of propylene glycol. Immunohistochemical evaluation of
Ki-67 marker was performed and checked carefully with microscopic analysis of the
cells of the vaginal epithelium, Statistical analysis was performed using the
nonparametric Mann Whitney test. Results: Light microscopy showed a higher
concentration of brown-stained cells with anti-Ki-67 in the control group than in the
experimental group. The mean percentage of nuclei stained with Ki-67 per 500 cells
in the vaginal epithelium was 68.64 ± 2.64 and 30.46 ± 2.00 [mean +/- standard error
of the mean (SEM)] in the control and (p <0.003). Conclusions: The present study
showed that anastrozole, in the dose and duration of treatment used, induced a
significant decrease in the cellular proliferation of the vaginal mucosa of rats in the
permanent estrus, evaluated by the expression of the protein Ki- 67.
