RESUMO:A função fisiológica do sistema imune é a defesa contra microrganismos infecciosos. Os linfócitos T realizam papel central na resposta imune adquirida e vem sendo cada vez mais alvo de estudo, devido aos seus mecanismos fisiológicos de auto-regulação. Nosso objetivo, neste trabalho, foi identificar o perfil molecular de mRNA em linfócitos T de moléculas envolvidas em imunorregulação (FOXP3, GATA3, TGF-β, IL-10,) e de outras envolvidas em resposta imune pró-inflamatória (T-bet, IFN-у, TNF- α), realizamos a interação de antígenos da Hsp60 com essas diferentes DCs em linfócitos, T no contexto de células dendríticas imaturas. Primeiramente Células dendríticas humanas maduras (mDC) e imaturas (iDC) foram geradas, in vitro, a partir de monócitos de 16 de indivíduos saudáveis. Estas células foram caracterizadas quanto à (i) morfologia e (ii) imunofentotipagem. Após a cocultura com antígenos da Hsp60, os linfócitos T foram purificados e feita a análise em RT- PCR. Obtivemos DC totalmente diferenciadas, a eficiência de diferenciação de monócitos em DC foi maior do que 90% após a purificação. As análises da morfologia revelaram que ambos os tipos celulares apresentam semelhanças morfológicas: são multiformes e sem vacúolos. No entanto, essas mesmas células apresentaram também algumas diferenças quanto à posição do núcleo e quanto às projeções membranosas. No perfil molecular verificamos um predomínio de modificações reguladoras para os genes TGF-β, TGF-β R, GATA3, TNF-α e T-bet. O peptídeo N7 induziu modificações dominantemente reguladoras em quase todas as condições em que ele foi testado. Em conclusão, verificamos que antígenos da Hsp60 têm a capacidade de ativar, simultaneamente, em linfócitos T, na interação com as células dendríticas, genes funcionalmente antagônicos. Isto reafirma a diversidade funcional da Hsp60. Ademais, identificamos o peptídeo N7 como potencialmente imunorregulador.
ABSTRACT:The physiological function of the system is a defense against infectious microorganisms. T lymphocytes play a central role in the immune response and are increasingly being studied because of their physiological mechanisms of self-regulation. Our objective was to determine the molecular profile of mRNA in T-lymphocytes involved in immunoregulation (FOXP3, GATA3, TGF-β, IL-10) and others involved in a proinflammatory immune response (T-bet, IFN-γ, TNF-α), we performed an interaction of Hsp60 antigens with these different DCs on T lymphocytes in the context of immature dendritic cells. First, mature human dendritic (mDC) and immature (dDC) cells were generated, in vitro, from monocytes of 16 from greetings. These cells were characterized for (i) morphology and (ii) immunofenthotyping. After coculture with Hsp60 antigens, the T lymphocytes were purified and analyzed in RT-PCR. We obtained fully differentiated DCs, a monocyte division efficiency in DC was greater than 90% after a clearance. As analyzes of the morphology revealed that both types of cellular are reassembled with morphologies: they are multiform and without vacuoles. However, however, these cells similar to some references to the position of the nucleus and to the membranous projections. In the molecular profile we observed a predominance of regulatory modifications for the genes TGF-β, TGF-β R, GATA3, TNF-α and T-bet. The N7 peptide induced dominant regulatory modifications in almost all conditions under which it was tested. In conclusion, we verified that Hsp60 antigens have the ability to simultaneously activate T lymphocytes in the interaction with dendritic cells, functionally antagonistic genes. This reaffirms a functional diversity of Hsp60. In addition, identifiers or peptide N7 as potentially immunoregulatory. |
