Abstract:
RESUMO:A diarreia é um dos distúrbios gastrintestinais que pode ser debilitante e quando não tratada pode
oferecer risco ao paciente, caracterizada pela eliminação excessiva de líquidos e sais do corpo na forma
de fezes líquidas. A terapia farmacológica atua de forma eficiente no controle do estado diarreico,
entretanto, os fármacos apresentam efeitos adversos que podem agravar o estado clínico. A Tocoyena
hispidula Standl. é conhecida popularmente como “flor-do-cerrado” e “angelca”, podendo ser localizada
nas regiões Norte e Nordeste. No Piauí, a Tocoyena hispidula Standl. é encontrada no município de
Campo Maior usada popularmente no tratamento de dor abdominal e inflamação do útero. Objetivouse
dessa forma avaliar a atividade antidiarreica do extrato etanólico das folhas de Tocoyena hispidula
Standl em camundongos. Inicialmente realizou-se a caracterização química do extrato entanólico Th-
EtOHf em HPLC, posteriormente realizou-se o teste de toxicidade aguda do extrato etanólico Th-EtOHf
conforme a OECD 423/2011. A atividade antidiarreica do extrato (Th-EtOHf) foi investigada no modelo
de diarreia aguda e enteropooling induzido por óleo de rícino em camundongos tratados com Th-EtOHf
(10; 50; 100 mg/kg, v.o.), salina (2,5 mL/kg, v.o.) ou loperamida (5 mg/kg, v.o.) e, 1 hora depois
receberam óleo de rícino (10 mL/kg, v.o.) e foram monitorados durante 4 horas. Logo após foram
analisadas a severidade diarreica, a massa total das fezes e a medida do volume do conteúdo intestinal.
Com o intuito de avaliar o efeito do Th-EtOHf sobre o trânsito intestinal em camundongo, os animais
receberam inicialmente o óleo de rícino e 1 hora depois, receberam o tratamento com Th-EtOHf (50
mg/kg, v.o). Decorrida mais 1 hora, receberam 0,2 mL de carvão ativado (v.o), sendo eutanasiados
após 20 min. Ao final, a distância percorrida pelo carvão foi então mensurada. O envolvimento das vias
opióide e muscarínica no trânsito foi avaliada, analisou-se também o desempenho do Th-EtOHf na
diarreia de cunho inflamatório (PGE2, 100 μg/kg, v.o.). Para isso os animais foram pré-tratados
oralmente com Th-EtOHf (50 mg/kg, v.o.), salina ou loperamida (5 mg/kg). Decorrido 30 min os animais
foram eutanasiados e o volume do conteúdo intestinal foi então analisado. O resultado da análise
química do extrato em HPLC revelaram bandas sugestivas de compostos da classe dos iridoides e
flavonoides, o extrato não demonstrou sinais de toxicidade aguda por via oral conforme os testes
preconizados nas doses avaliadas apresentaram um efeito significativo (*p< 0,05), diminuindo a
quantidade total de fezes em 38,07%; 50,33%; 52,98% (respectivamente) e fezes diarreicas em
30,52%; 46,98%; 46,18% apresentando também valores significativos no protocolo de enteropooling,
com melhores resultados na dose de 50 mg/kg, permanecendo esta como padrão para os demais
testes. O Th-EtOHf não exerceu efeitos significativos no modelo de diarreia induzida por PGE2. O Th-
EtOHf reduziu o trânsito gastrintestinal, através da via muscarínica, sendo capaz de diminuir
significativamente em 43,07% a quantidade total de fezes e reduzir a produção de fluidos intestinais.
Diante dos resultados expostos, foi possível constatar a atividade antidiarreica do Th-EtOHf, mostrandoser um potencial insumo, para produção de medicamentos, para o tratamento de doenças diarreicas.ABSTRACT:Diarrhea is one of the gastrointestinal disorders that can be debilitating and when left untreated may

antidiarrheal activity of the ethanolic extract from the leaves of Tocoyena hispidula Standl in mice. nitially

induced by castor oil in mice treated with Th-EtOHf (10, 50, 100 mg / kg, / kg) or loperamide (5 mg / kg)
and then 1 hour later received castor oil (10 ml / kg, vol) monitored for 4 hours. Soon after, the diarrheal
severity, the total weight of the feces and the measurement of the volume of the intestinal contents were
analyzed. In order to evaluate the effect of Th-EtOHf on intestinal transit in mice, the animals initially
received castor oil and 1 hour later were treated with Th-EtOHf (50 mg / kg, v.o). After another 1 hour,
they received 0.2 mL of activated charcoal (v.o) and were euthanized after 20 min. At the end, the
distance traveled by the coal was then measured. The involvement of the opioid and muscarinic
pathways in the traffic was evaluated, the performance of Th-EtOHf in inflammatory diarrhea (PGE2, 100
μg / kg, v.o.) was also investigated. For this the animals were orally pretreated with Th-EtOHf (50 mg /
kg, v.o.), saline or loperamide (5 mg / kg). After 30 min the animals were euthanized and the volume of
the intestinal contents was analyzed.The results of HPLC extract analysis revealed bands suggestive of
compounds of the class of iridoids and flavonoids, the extract did not demonstrate acute toxicity
according to The antidiarrheal effects in the doses evaluated had a significant effect (* p <0.05), reducing
the total amount of feces in 38.07%; 50.33%; 52.98% (respectively) and diarrheal stools in 30.52%;

effect on the PGE2-induced diarrhea model. Th-EtOHf reduced the gastrointestinal 43.07% transit
through the muscarinic pathway, being able to significantly decrease the total amount of feces and
reduce the production of intestinal fluids. In view of the aforementioned results, it was possible to verify
the antidiarrheal activity of Th-EtOHf, proving to be a potential therapeutic input for the treatment of
diarrheal diseases.
