RESUMO: A dor é um dos sintomas mais relatados em serviços de saúde. Contudo, as opções farmacológicas para seu tratamento costumam provocar frequentes eventos adversos, tornando o tratamento intolerável para muitos pacientes. Portanto, a fim de ampliar as possibilidades de tratamento, são realizados estudos que buscam novos fármacos, estando os produtos naturais incluídos. O terpinoleno (TPO) é um monoterpeno que compõe óleos essenciais de plantas frequentemente encontradas no nordeste brasileiro, como Chenopodium ambrosioides L. (mastruz), Citrus aurantium L. (laranja amarga) e Cumminum cyminum L. (cominho), com algumas propriedades terapêuticas já sugeridas. Assim, esta tese se fundamentou em quatro capítulos, com os seguintes objetivos: o capítulo I consistiu em realizar uma revisão de literatura científica descrevendo a fisiopatologia da dor neuropática, bem como os tratamentos convencionalmente utilizados, e novas perspectivas terapêuticas, envolvendo o TPO como possibilidade. No capítulo II, objetivou-se elaborar uma outra revisão de literatura científica, abordando as classes de fármacos que poderiam gerar dor neuropática como efeito adverso, com a descrição dos respectivos mecanismos fisiopatológicos. No capítulo III, consistiu-se em elaborar formulações transdérmicas contendo TPO, por meio de um sistema de microemulsão detalhadamente descrito. No capítulo IV, foi testada a ação no TPO por via oral e transdérmica, por meio das formulações desenvolvidas no capítulo III, sobre o modelo de neuropatia por ligadura do nervo ciático. O efeito sobre a atividade locomotora dos camundongos também foi testado (teste do campo aberto e Rota rod), bem como sobre os níveis de malondialdeído, glutationa reduzida e sobre a atividade da superóxido dismutase. Os resultados demonstraram que o método selecionado para desenvolvimento do sistema de microemulsão para as formulações transdérmicas de TPO, no capítulo III, foi satisfatório, o que permitiu a obtenção das formulações F4 e F8, que apresentaram os melhores resultados no teste da cinética de liberação, sendo essas utilizados para os testes in vivo do capítulo IV. Além disso, o TPO, tanto livre (100 e 200 mg/kg vo) quanto transdérmico, sugeriu efeito antinociceptivo nos testes de von Frey e acetona, tanto com administrações agudas quanto subagudas (por 8 dias), sem, contudo, alterar a atividade locomotora no teste do campo aberto no Rota rod. Esse efeito não esteve correlacionado aos níveis de malondialdeído glutationa reduzida e nem atividade da superóxido dismutase. ABSTRACT: Pain is one of the most reported symptoms in health services. However, the pharmacological options for its treatment usually cause frequent adverse events, making the treatment intolerable for many patients. Therefore, in order to expand the possibilities of treatment, studies are conducted with new options of drugs, with natural products. Terpinolene (TPO) is a monoterpene wich composes essential oils of plants frequently found in northeastern Brazil, such as Chenopodium ambrosioides L. (mastruz), Citrus aurantium L. (bitter orange) and Cumminum cyminum L. (cumin), with some therapeutic properties suggested. Thus, this thesis was based on four chapters, with the following objectives: Chapter I consisted of a review of scientific literature describing the pathophysiology of neuropathic pain, as well as the treatments conventionally used, and new therapeutic perspectives, involving TPO as a possibility. Chapter II aimed to elaborate another review of scientific literature, addressing the classes of drugs wich could generate neuropathic pain as an adverse effect, with a description of the respective pathophysiological mechanisms. Chapter III consisted of elaborate transdermal formulations containing TPO by means of a detailed microemulsion system. In Chapter IV, oral and transdermal TPO action was tested through formulations developed in Chapter III on the sciatic nerve ligation neuropathy model. The effect on the locomotor activity of the mice was also tested (open field test and Rota rod), as well as levels of malondialdehyde, reduced glutathione and superoxide dismutase activity. The results showed that the method selected for the microemulsion system for TPO transdermal formulations in chapter III was satisfactory, which allowed to select F4 and F8 formulations, which presented the best results in the release kinetics test, being used for the in vivo tests of Chapter IV. In addition, TPO, both free (100 and 200 mg/kg, orally) and transdermal, suggested an antinociceptive effect in the von Frey and acetone tests, both with acute and subacute administrations (for 8 days), without, however, changing the locomotor activity in the open field test and Rota rod test. This effect was not correlated with malondialdehyde and reduced glutathione levels nor superoxide dismutase activity. |
