Abstract:
RESUMO: O Borneol é um monoterpeno bicíclico que pode ser obtido como mistura racêmica [(+)-Borneol + (-) - Borneol] a partir de óleos essenciais de plantas medicinais como a Valeriana officinalis L. (Erva de gato), Matricaria chamomilla L. (Camomila), Lavandula officinalis Chaix & Kitt (Alfazema) ou como molécula sintética de grande aplicação na indústria alimentícia, cosmética e farmacêutica. Estudos farmacológicos do Borneol indicam atividade cardiovascular, aumento da permeabilidade da barreira hematoencefálica, antinocicepção e neuroproteção. Este trabalho teve o objetivo de investigar o efeito ansiolítico e antiadictivo do (-) - Borneol [(-) – BOR] sintético com o intuito de contribuir com maiores informações farmacológicas e toxicológicas sobre este monoterpeno, visando um embasamento maior da molécula como uma alternativa terapêutica no tratamento da ansiedade e da adicção. Os resultados obtidos através desse estudo foram apresentados em 3 (três) capítulos. No primeiro capítulo investigou-se o possível efeito sedativo e ansiolítico do (-) - BOR em camundongos. O (-) - BOR apresentou efeito ansiolítico ao reduzir o tempo de início e prolongar o tempo total de sono induzido por tiopental sódico. Além disso, o (-) - BOR também apresentou efeito ansiolítico ao reduzir a atividade locomotora dos animais no teste do campo aberto e ao aumentar o tempo de permanência dos animais na área clara e nos braços abertos nos testes da caixa claro-escuro e labirinto em cruz elevado, respectivamente. Como embasamento teórico, realizou-se um estudo de docagem molecular entre o (-) - BOR e o receptor GABAA (GABAAR). Assim, no estudo de docagem, foi possível perceber que existe interação molecular entre o monoterpeno e o GABAAR sendo este, portanto, um possível mecanismo de ação ansiolítica. No segundo capítulo investigou-se a atividade do (-) - BOR em camundongos dependentes e abstinentes a morfina. No estudo, foi possível observar que o (-) - BOR interfere no desenvolvimento de preferência condicionada por lugar (PCL) por morfina ao reduzir o tempo de permanência dos animais no ambiente pareado e na abstinência, ao reduzir o comportamento de saltar. A avaliação do efeito genotóxico do (-) - BOR foi realizada em sangue periférico dos camundongos através do teste do cometa. Em nenhum dos parâmetros observados o (-) - BOR apresentou efeito genotóxico. No terceiro capítulo, investigou-se o efeito neuroprotetor do (-) - BOR na fragmentação de DNA induzida por morfina em áreas mesolímbicas como o hipocampo e córtex pré-frontal de camundongos. O (-) - BOR reduz o dano ao DNA induzido por morfina nas duas áreas mesolímbicas avaliadas, o que provavelmente protegeria essas áreas em pacientes que utilizam morfina de forma crônica. A avaliação do (-) - BOR sobre a atividade da enzima lactato desidrogenase (LDH) demonstra que os efeitos do monoterpeno não podem ser relacionados com atividade antinecrótica, pois tanto o (-) - BOR quanto a morfina não interferem na atividade da enzima. Assim, investigação do (-) - BOR revelou que a molécula possui propriedades ansiolíticas, antiadictivas e neuroprotetora observadas em modelos animais com camundongos Swiss webster. ------------------ ABSTRACT: Borneol is a bicyclic monoterpene which can be obtained as a racemic mixture [(+) - Borneol + (-) - Borneol] from essential oils of medicinal plants such as Valeriana officinalis L. (Cat's grass), Matricaria chamomilla L. (Chamomile), Lavandula officinalis Chaix & Kitt (Lavender) or as synthetic molecule of great application in the food, cosmetic and pharmaceutical industry. Pharmacological studies of Borneol point to cardiovascular activity, increased permeability of the blood-brain barrier, antinociception and neuroprotection. This work aimed to investigate the anxiolytic and antiaddictive effect of synthetic (-) - Borneol [(-) - BOR] in order to contribute with greater pharmacological and toxicological information about this monoterpene, aiming a greater base of the molecule as therapeutic alternative in treatment of anxiety and Addiction. The results obtained through this study were presented in 3 (three) chapters. In the first chapter, we investigated the possible sedative and anxiolytic effect of (-) - BOR in mice. (-) - BOR presented anxiolytic effect by reducing the onset time and prolonging the total sleep time induced by thiopental sodium. In addition, (-) - BOR also presented an anxiolytic effect by reducing the locomotor activity of the animals in the open field test and increasing the spent time of the animals both in the light area and in the open arms in the tests of the light-dark box and elevated plus maze, respectively. As a theoretical basis, a molecular docking study was performed between the (-) - BOR and the GABAA receptor (GABAAR). Thus, in the docking study, it was possible to observed that there is a molecular interaction between monoterpene and GABAAR, which is, therefore, a possible mechanism of anxiolytic action. In the second chapter the activity of (-) - BOR was investigated in morphine-dependent and abstinent mice. In the study, it was possible to observe that (-) - BOR interferes in the development of place-based preference (PCP) for morphine by reducing the spent time of the animals in the paired environment and in abstinence by reducing jumping behavior. The evaluation of the genotoxic effect of (-) - BOR was performed in peripheral blood of the mice through the comet assay. In none of the parameters observed did (-) - BOR show genotoxic effect. In the third chapter, we investigated the neuroprotective effect of (-) - BOR on morphine-induced DNA fragmentation in mesolimbic areas such as the hippocampus and prefrontal cortex of mice. (-) - BOR reduces morphine-induced DNA damage in the two mesolimbic areas evaluated, which would probably protect these areas in patients using chronic morphine. The evaluation of (-) - BOR on the activity of the enzyme lactate dehydrogenase (LDH) demonstrates that the effects of monoterpene can not be related to antinecrotic activity, since both (-) - BOR and morphine do not interfere in enzyme activity. Thus, investigation of (-) - BOR revealed that the molecule has anxiolytic, anti-addictive and neuroprotective properties observed in animal models with Swiss webster mice.
