CARACTERIZAÇÃO FISIOFARMACOLÓGICA IN VITRO E IN VIVO DE NOVOS DERIVADOS DA CLASSE DAS 2-CLORO-N-ARILACETAMIDAS: potencial antineoplásico e aspectos toxicológicos

Abstract

RESUMO: As estimativas para os anos de 2018/2019 no Brasil será cerca de 600 mil novos casos de câncer, no entanto o tratamento quimioterápico leva a efeitos colaterais, assim pesquisas de novas moléculas que proporcionem efeitos antitumorais com seletividade em relação a toxicidade de células normais. O presente estudo avaliou as atividades citotóxicas, tóxicas e antitumorais de compostos sintéticos 2-cloro-N-4-clorofenil acetamida (composto 2), 3: 2-cloro-N-4-bromofenil acetamida (composto 3) e 2-cloro-N-4-nitrofenil acetamida (composto 4), derivados da classe 2-cloro-N-arilacetamidas em estudos in vitro e in vivo. Os compostos avaliados foram escolhidos após triagem de atividades antiproliferativas de dez (1-10) derivados sintéticos de 2-cloro-N-arilacetamidas, em linhagens tumorais (HL-60: leucemia promielocítica, HCT-116: carcinoma colorretal, SF-295: glioblastoma, HEPG-2: carcinoma de fígado e OVCAR-8: carcinoma de ovário) e em culturas primárias de células mononucleares do sangue periférico, bem como atividade hemolítica em eritrócitos humanos. Os compostos 2 (2-cloro-N-4-clorofenil acetamida), 3 (2-cloro-N-4-bromofenil acetamida) e 4 ( 2-cloro-N-4-nitrofenil acetamida) em concentrações crescentes (0,04-25 µg/mL) foram analisados quanto sua citotoxicidade em linhagens celulares de fibroblastos normais (MRC-5: humano e L-929: murino). A toxicidade dos compostos foi avaliada em meristema de Allium cepa, em Artemia salina e em embriões de peixe-zebra (FET) com o composto 2. Em Saccharomyces cerevisiae foi avaliado a atividade antioxidante/oxidante. Em camundongos nas doses de 150 e 300 mg/kg foram avaliados quanto aos aspectos toxicológicos e comportamentais. Atividade antitumoral dos compostos na dose de 25 mg/kg/dia foi avaliada em camundongos imunodeficientes transplantados com células de carcinoma de cólon. Os compostos 2, 3 e 4 apresentaram valores de CI50 frente às linhagens tumorais variando de 1 a >10 µg/mL, de 4,2 a 9,8 µg/mL e 2,4 a 8,9 µg/mL, respectivamente. Os compostos avaliados não foram citotóxicos em eritrócitos e células mononucleares do sangue periférico humanas. Em A. salina os três compostos apresentaram baixa CE50 (composto 2: 0,005 µg/mL; composto 3: 0,009 µg/mL e composto 4: 7,06 µg/mL), portanto sendo classificados como tóxicos. Em embriões de peixe-zebra tratados com o composto 2 nas concentrações de 1,93 e 7,2 mg/L, apresentaram percentuais de 90% e 100% de morte. Na avaliação da atividade citotóxica em células de fibroblasto humano e murino os compostos apresentaram CI50 de 2,5; 4,8 e 1,2 µg/mL para os compostos 2, 3 e 4 respectivamente. Como também em meristemas de raízes de Allium cepa por inibição do índice mitótico nas concentrações 1, 5, 10, 25 e 50 µg/mL, apresentando efeitos genotóxicos. Em Saccharomyces cerevisiae o composto 1 não interfere em danos oxidativos causados pelo peróxido de hidrogênio, entretanto os compostos 2 e 3 apresentaram atividades antioxidantes nas menores concentrações testadas (1 e 5 µg/mL). Na avaliação da toxicidade em camundongos, foram observadas mortes somente na dose de 300 mg/kg, não apresentando alterações exploratórias e motoras. Na investigação do potencial antitumoral in vivo, apenas os compostos 1 e 2 reduziram o crescimento tumoral em 21,2% e 27,5% quando comparados ao controle negativo. O estudo aponta que os compostos 1, 2 e 3 apresentaram potencial antitumoral in vitro e in vivo, com seletividade em relação às células normais humanas, como também não apresentaram toxicidade por alterações hipocráticas e comportamentais em animais. Assim sugere-se que os compostos avaliados podem ser usados em formulações farmacêuticas, como antitumorais. ABSTRACT: The estimates for the years 2018/2019 in Brazil will be about 600 thousand new cases of cancer, however the chemotherapy treatment leads to side effects, thus research of new molecules that provide antitumor effects with selectivity in relation to normal cell toxicity. The present study evaluated the cytotoxic, toxic and antitumor activities of synthetic compounds 2-chloro-N-4-chlorophenylacetamide (compound 2), 3: 2-chloro-N-4-bromophenylacetamide (compound 3) and 2-chloro- N-4-nitrophenylacetamide (compound 4), derivatives of class 2-chloro-N-arylacetamides in in vitro and in vivo studies. The compounds evaluated were selected after screening for antiproliferative activities of ten (1-10) synthetic 2-chloro-N-arylacetamide derivatives in tumoral lines (HL-60: promyelocytic leukemia, HCT-116: colorectal carcinoma, SF-295: glioblastoma, HEPG-2: liver carcinoma and OVCAR-8: ovarian carcinoma) and in primary cultures of peripheral blood mononuclear cells, as well as hemolytic activity in human erythrocytes. The compounds 2- (2-chloro-N-4-chlorophenylacetamide), 3-(2-chloro-N-4-bromophenylacetamide) and 4- (2-chloro-N-4-nitrophenylacetamide) in increasing concentrations -25 μg / ml) were analyzed for cytotoxicity in normal fibroblast cell lines (MRC-5: human and L-929: murine). The toxicity of the compounds was evaluated in Allium cepa meristem, Artemia salina and zebrafish (FET) embryos with compound 2. In Saccharomyces cerevisiae the antioxidant / oxidant activity was evaluated. In mice at doses of 150 and 300 mg / kg were evaluated for toxicological and behavioral aspects. Antitumor activity of the compounds at the dose of 25 mg / kg / day was evaluated in immunodeficient mice transplanted with colon carcinoma cells. Compounds 2, 3 and 4 presented IC50 values against tumoral strains ranging from 1 to> 10 μg / mL, from 4.2 to 9.8 μg / mL and 2.4 to 8.9 μg / mL, respectively. The evaluated compounds were not cytotoxic to erythrocytes and human peripheral blood mononuclear cells. In all three compounds, the three compounds showed low EC 50 (compound 2: 0.005 μg / mL, compound 3: 0.009 μg / mL and compound 4: 7.06 μg / mL), thus being classified as toxic. In zebrafish embryos treated with compound 2 at concentrations of 1.93 and 7.2 mg / L, they presented 90% and 100% percentages of death. In the evaluation of cytotoxic activity in human and murine fibroblast cells the compounds had IC 50 of 2.5; 4.8 and 1.2 μg / mL for compounds 2,3 and 4 respectively. As well as in root meristem of Allium cepa by inhibition of the mitotic index at concentrations 1, 5, 10, 25 and 50 μg / mL, presenting genotoxic effects. In Saccharomyces cerevisiae, compound 1 does not interfere with oxidative damage caused by hydrogen peroxide. However, compounds 2 and 3 presented antioxidant activities at the lowest concentrations tested (1 and 5 μg / mL). In the evaluation of toxicity in mice, deaths were observed only at the dose of 300 mg / kg, without exploratory and motor alterations. In the investigation of the antitumor potential in vivo, only compounds 1 and 2 reduced tumor growth by 21.2% and 27.5% when compared to the negative control. The study shows that compounds 1, 2 and 3 showed antitumor potential in vitro and in vivo, with selectivity in relation to normal human cells, nor did they present toxicity due to hipocrátic and behavioral alterations in animals. Thus it is suggested that the evaluated compounds can be used in pharmaceutical formulations, such as antitumor.
