RESUMO: As substâncias encontradas no gengibre (Zingiber officinale Roscoe) apresentam ações antitumoral, antimutagênica, antioxidante e anti-inflamatória. O estudo avaliou os efeitos e mecanismos toxicogenéticos e antitumorais do [6]-Gingerol ((S)-5-hidroxi-1-(4-hidroxi-3-metoxifenil)-3-decanona) em ensaios não clínicos. A toxicidade e citotoxicidade foi avaliada em Artemia salina, Allium cepa e MTT (brometo de 3-[4,5-dimetil-tiazol-2-il]-2,5-difenil tetrazólio). Efeitos antitumorais foram avaliados em linhagem humana para leucemia, murinos com Sarcoma 180 e carcinoma mamário, induzido pelo 7,12-dimetilbenzantraceno, com aplicação de marcadores citogenéticos. O [6]-Gingerol foi testado em 5, 10, 20, 40, 60, 80 e 100 μg.mL-1. O [6]-Gingerol apresentou toxicidade e citotoxicidade a partir de 10 μg.mL-1 em Artemia salina e Allium cepa, apresentando concentração inibitória no ensaio MTT de 1,14; 5,73 e de 11,18 μg.mL-1, em linhagem leucêmica, Sarcoma 180 e em células mononucleadas de sangue periférico, respectivamente. Nas concentrações de 10 e 20 μg.mL-1, o [6]-Gingerol apresentou efeitos antitumorais em Sarcoma 180 pela redução de viabilidade celular e do índice de divisão celular, por mecanismos genotóxicos similares aos danos oxidativos induzidos pelo H2O2. Efeitos antitumorais do [6]-Gingerol em 10 mg.Kg-1, também foram observados em camundongos fêmeas com carcinoma mamário, evidenciado pelas análises histopatológicas e redução da marcação da proteína Ki-67. O tratamento com [6]-Gingerol não demonstrou alterações na avaliação comportamental dos animais, o perfil hematológico apontou conformidade na série vermelha, porém, aumento de eosinófilos, na série branca. Na avaliação bioquímica para enzimas hepáticas, foi observado aumento significativo de alanina aminotransferase, contudo, verificou-se ausência de alterações renais (ureia e creatinina). O [6]-Gingerol induziu baixa toxicidade, genotoxicidade e apoptose em células não tumorais (sangue periférico, medula óssea e fígado) quando comparado a ciclofosfamida (25 mg.kg-1). Por outro lado, estes efeitos foram proeminentes em tecido neoplásico. Assim, o [6]-Gingerol teve efeitos antitumorais por mecanismos citogenéticos indicativos de citotoxicidade e genotoxicidade por danos oxidativos, mutagenicidade pela formação de brotos e pontes nucleoplásmicas, que culminaram para a indução de apoptoses. Os efeitos observados em estudos não clínicos podem contribuir para novas análises necessárias para o uso do [6]-Gingerol em formulações farmacêuticas antitumorais.
ABSTRACT: Substances found in ginger (Zingiber officinale Roscoe) have showed anti-tumor, antimutagenic, antioxidant and anti-inflammatory properties. This study evaluated the toxicogenetic and antitumoral effects of [6]-Gingerol ((S)-5-hydroxy-1-(4-hydroxy-3-methoxyphenyl)-3-decanone), as well as mechanisms related to these effects in non-clinical trials. Toxicity and cytotoxicity was evaluated in Artemia salina, Allium cepa and MTT (3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyl bromide) assays, while toxicogenetic and antitumoral effects were evaluated in HL-60, Sarcoma-180 and 7,12-dimethylbenzanthracene-induced mammary carcinoma in mice, with application of cytogenetic markers. [6]-Gingerol was tested at 5, 10, 20, 40, 60, 80 and 100 μg.mL-1. The compound showed toxicity and cytotoxicity from 10 μg.mL-1 in Artemia salina and Allium cepa, with an inhibitory concentration of 1,14; 5,73 and 11,18 μg.mL-1, in HL-60, Sarcoma-180 and in peripheral blood mononuclear cells, respectively, in MTT. At 10 and 20 μg.mL-1 we observed antitumoral effects in Sarcoma-180 by reduction of cell viability and cell division index, due to genotoxic mechanisms similar to oxidative damage induced by H2O2. Anti-tumor effects of [6]-Gingerol in 10 mg.Kg-1 were also observed in female mice with mammary carcinoma, evidenced by histopathological analyzes and reduced labeling for Ki-67. The treatment with [6]-Gingerol did not show alterations in the behavior of the animals. Moreover, the hematological profile showed compliance in the red series, although it has been evidenced an increase in eosinophils in the white series. Also, an increase of alanine aminotransferase was observed after therapy with [6]-Gingerol. Interestingly, [6]-Gingerol induced low toxicity, genotoxicity and apoptosis in non-tumor cells (peripheral blood, bone marrow and liver cells) when compared to therapy with cyclophosphamide (25 mg.kg-1). But these effects were prominent in breast carcinoma. Thus, [6]-Gingerol had anti-tumor effects by cytogenetic mechanisms indicative of cytotoxicity and genotoxicity by oxidative damage, mutagenicity by the formation of nucleoplasmic bridges and buds, which culminated in the induction of apoptosis. These effects observed in non-clinical studies may contribute to further analyzes necessary for the use of [6]-Gingerol in pharmaceutical formulations with emphasis on the oncological context. |
