Abstract:
RESUMO:S. acuruensis (Benth.) (Fabaceae) é uma espécie endêmica do nordeste brasileiro, com
ocorrência no estado do Piauí, conhecida popularmente como canafistinha e besouro. O
presente trabalho relata os constituintes químicos isolados e identificados no extrato
etanólico das cascas dos frutos (EEC) desta espécie, bem como o potencial biológico do
extrato, frações e compostos isolados. O EEC foi submetido à cromatografia em coluna
filtrante de gel de sílica, fornecendo as frações hexânica (FH), clorofórmica (FCHCl3),
AcOEt (FAcOEt) e metanólica (FMeOH). As frações FH, FCHCl3 e FAcOEt foram
submetidas a cromatografia em coluna de gel de sílica e/ou Sephadex LH-20. A
investigação fitoquímica do EEC resultou no isolamento e caracterização de uma nova
antraquinona, 9,10-dioxo-1-formil-3-metil-[5,6,1’,2’]-furanoantraquinona (1), como
também nos compostos conhecidos 1,8-dihidroxi-3-metil-6-metoxiantraquinona (2),
lupeol (3), α-amirina (4), β-amirina (5), germanicol (6), campesterol (7), sitosterol (8),
estigmasterol (9), 7α-hidroxisitosterol (10), 7β-hidroxisitosterol (11), 7α-
hidroxiestigmasterol (12), Δ22-estigmastenol (13), 6β-hidroxistigmast-4-en-3-ona (14),
6β-hidroxiestigmasta-4,22-dien-3-ona (15), peróxido de ergosterol (16), fomentarol C
(17), 6-O-acil-β-D-glicosil-3-O-sitosterol (18), 6-O-acil-β-D-glicosil-3-O-estigmasterol
(19), (E)-fitol (20), (Z)-fitol (21), 11,12,13-trimetilhexil-3-metilhexa-3,5-dienoato (22),
hexatriacontano (24) e ácidos graxos saturados e insaturados (23a-23g). As estruturas
desses compostos foram identificadas e/ou elucidadas utilizando técnicas
espectrométricas, como RMN 1H e 13C (1D e 2D), IV, ESI-MS, CG-EM e por
comparação com dados relatados na literatura. O EEC, as frações FCHCl3, FAcOEt e
FMeOH apresentaram inibição da enzima acetilcolinesterase. O EEC, FAcOEt e 1
inibiram o crescimento de promastigotas de Leishmania major, com valores de
concentação inibitória média (CI50) de 68,8, 36,7 e 11,2 µg mL-1, respectivamente. Este
é o primeiro relato do composto 1 na literatura, 10-15 no gênero Senna, 16 na família
Fabaceae, 17 e 22 em plantas, bem como, da ação antileishmania na espécie.
ABSTRACT:S. acuruensis (Benth.) (Fabaceae) is an endemic species from brazilian northeast,
occurring in the state of Piauí, popularly known as canapistinha and beetle. The present
work reports the chemical constituents isolated and identified in the ethanolic extract of
fruit peels (EEC) from this species, as well as the biological potential of the extract,
fractions and isolated compounds. The EEC was subjected to a filtration silica gel
column chromatography, providing the hexane (FH), chloroform (FCHCl3), AcOEt
(FAcOEt) and methanolic (FMeOH) fractions. The FH, FCHCl3 and FAcOEt fractions
were subjected to silica gel column chromatography and/or Sephadex LH-20. The
phytochemical investigation of EEC resulted in the isolation and characterization of a
novel anthraquinone, 9,10-dioxo-1-formyl-3-methyl-[5,6,1', 2']-furananthraquinone (1),
as well as the known compounds 1,8-dihydroxy-3-methyl-6-methoxyanthraquinone (2),
lupeol (3), α-amyrin (4), β-amyrin (5), germanicol (6), campesterol (7), sitosterol (8),
stigmasterol (9), 7α-hydroxysitosterol (10), 7β-hydroxysitosterol (11), 7α-
hydroxystigmasterol (12), Δ22-stigmastenol (13), 6β-hydroxystygmast-4-en-3-one (14),
6β-hydroxystigmasta-4,22-dien-3-one (15), ergosterol peroxide (16), fomentarol C (17),
6-O-acyl-β-D-glycosyl-3-O-sitosterol (18), 6-O-acyl-β-D-glycosyl-3-O-stigmasterol
(19), (E)-phytol (20), (Z)-phytol (21), 11,12,13-trimethylhexyl-3-methylhexa-3,5-
dienoate (22), hexatriacontane (24) saturated and unsaturated fatty acids (23a-23g).
Structures of these compounds have been identified and/or elucidated using
spectrometric techniques, such as NMR 1H and 13C (1D and 2D), IV, ESI-MS, GC-MS
and by comparison with data reported in the literature. The EEC, the fractions FCHCl3,
FAcOEt and FMeOH showed inhibition of the enzyme acetylcholinesterase. The EEC,
FAcOEt and 1 inhibited the growth of Leishmania major promastigotes, with mean
inhibitory concentration (IC50) values of 68.8, 36.7 and 11.2 µg mL-1, respectively. This
is the first report of compound 1 in the literature, 10-15 in the genus Senna, 16 in the
Fabaceae family, 17 and 22 in plants, as well as the antileishmanial activity in the
species.
