RESUMO
A utilização de polissacarídeos derivados de gomas naturais é uma alternativa para sistemas drug delivey, devido as suas propriedades biocompatíveis, o que resulta em melhorias nos efeitos adversos dos fármacos. Esse trabalho estudou a utilização de nanopartículas da goma de angico branco purificada (NpAB) e do angico branco misturada com glicerol (NpAGL) como entregadores de Anfotericina B. As
nanopartículas foram sintetizadas, diálisada e caracterizadas por microscopia eletrônica de varredura (MEV), espectroscopia na região do infravermelho com transformada de Fourrier (IV), ressonância magnética nuclear (RMN),
termogravimetria (TG), quanto a sua distribuição de tamanho médio e potencial zeta,
quantidade de fármaco incorporado e liberado e atividade hemolítica. A goma de angico foi isolada do exsudado com rendimento de 87,54 % e as etapas de purificações com 74,67 e 78,93 %. Os espectros das nanopartículas carregadas com AmB absorveram na região do IV em 1100 e 950 cm-1 caracterizando a incorporação
da Anfotericina B. As análises termogramimétricas auxiliaram na caracterização, ficando evidente a incorporação do fármaco, a presença de heteropolissacarídeo na goma de angico branco e total polimerização do glicerol na formação da NpAGL. As
nanopartículas NpAB apresentaram tamanhos entre 45,1 e 267,6 nm e NpAGL apresentaram tamanhos variados de 49,3 nm, 176,2 nm e 828,0 nm, após a incorporação do fármaco, as NpAB-AmB apresentaram tamanho 131,7 nm e as
NpAGL-AmB tiveram a formação de tamanho de 53,3 nm e 161,9 nm. As imagens de MEV mostraram nanopartículas esféricas. O estudo de liberação in vitro do fármaco seguiu de liberação controlada até aproximadamente 24 h, e pôde-se verificar que este se ajusta ao modelo matemático Log-Logistic. As nanopartículas não apresentaram característica hemolítica.
ABSTRACT
The use of polysaccharides derived from natural gums is an alternative for drug delivey systems, due to their biocompatible properties, which results in improvements in the adverse effects of the drugs. This work investigated the use of nanoparticles of purified white angico gum (NpAB) and white angico mixed with glycerol (NpAGL) as amphotericin B delivery agents. The nanoparticles were synthesized, dialyzed and characterized by scanning electron microscopy (SEM), spectroscopy in the infrared region (IR), nuclear magnetic resonance (NMR), thermogravimetry (TG), as well as their distribution of medium size and zeta potential, amount of incorporated and released drug and hemolytic activity. Angico gum was isolated from the exudate in 87,54 % yield and the purification steps were 74,67 and 78,93 %. The spectra of the nanoparticles loaded with AmB absorb in the IR region 1100 and 950 cm-1 featuring the incorporation of amphotericin B. Thermograms helped in the characterization, making evident the incorporation of the drug, the presence of heteropolysaccharide in the white angico gum and total polymerization of glycerol in the formation of NpAGL.
The NpAB nanoparticles presented sizes between 45,1 and 267,6 nm and NpAGL
presented varying sizes of 49,3 nm, 176,2 nm and 828,0 nm, after incorporation of the drug, NpAB-AmB presented a size 131,7 nm and NpAGL-AmB had the size formation of 53,3 nm and 161,9 nm. MEV images showed spherical nanoparticles. The in vitro release study of the drug followed controlled release until approximately 24 h, and it
