Abstract:
RESUMO: A Medicina Regenerativa tem evoluído nos últimos anos e poderá ser de grande benefício para os pacientes como as que apresentam estrias, uma condição
desfigurante e aestética da pele, com opções terapêuticas gerenciadas sem reversão
efetiva. A regulamentação da técnica e da ética está se tornando mais clara facilitando a ciência a evoluir nas pesquisas com células tronco na restauração de tecidos, utilizando técnicas menos invasivas. Objetivo: O estudo tem como objetivo principal expandir e caracterizar células tronco mesenquimais originadas da Geleia de Wharton e da Placenta
que poderão ser utilizadas para restaurar a pele lesionada por estrias. Método: As células isoladas do cordão umbilical e da placenta foram expandidas e caracterizadas seguindo o protocolo para diferentes estágios de identificação das características morfofuncionais
e de imunofenotipagem. Resultados: No experimento as células da Geleia de Wharton cumpriram todas as etapas do nosso objetivo, embora as células da Placa Coriônica e da Vilosidade Coriônica não tenham completado todas as etapas, nenhum dos tipos de células cultivadas demonstrou características adversas às de células tronco. Discussão: O termo expansão celular é utilizado para a remoção de células de tecidos ou órgãos em
um ambiente artificial propício à sua sobrevivência e proliferação. Os requisitos ambientais básicos para que as células cresçam são: temperatura controlada, substrato para adesão celular, meio de crescimento adequado e incubadora que mantém o pH. As
mesmas vias metabólicas e bioquímicas de uma célula no organismo são consideradas
nas células em expansão in vitro. Conclusão: Os resultados mostraram a necessidade de padronização de protocolos para cada tipo de células de origem específica. Dentro do protocolo utilizado, as células tronco provenientes da Geleia de Wharton mostraram ser
mais viáveis à restauração da pele. ABSTRACT: Regenerative Medicine has evolved in recent years and may be of great benefit to patients such as those with stretch marks, a disfiguring condition and aesthetics of the skin, with therapeutic options managed without effective reversion. The regulation of technique and ethics is becoming clearer by facilitating science to evolve in stem cell research in tissue restoration using less invasive techniques. Objective: The main goal of the study is to expand and characterize mesenchymal stem cells originated from Wharton Jelly and Placenta that can be used to restore skin damaged by stretch marks. Methods: The cells isolated from the umbilical cord and the placenta were expanded and characterized following the protocol for different stages of identification of morphofunctional characteristics and immunophenotyping. Results: In the experiment Wharton Jelly cells fulfilled all the steps of our goal, although the cells of the Chorionic Plate and Chorionic Villus did not complete all the steps, none of the types of cells cultured demonstrated characteristics adverse to those of stem cells. Discussion: The term cell expansion is used to remove cells from tissues or organs in an artificial environment conducive to their survival and proliferation. The basic environmental requirements for cells to grow are: temperature controlled, substrate for cell adhesion, suitable growth medium and incubator that maintains the pH. The same metabolic and biochemical pathways of a cell in the body are considered in expanding cells in vitro. Conclusion: The results showed the need for standardization of protocols for each cell type of specific origin. Within the protocol used, stem cells from Wharton Jelly were shown to be more viable to restore skin.
