AVALIAÇÃO IN VITRO DO SÊMEN CAPRINO CRIOPRESERVADO EM DILUENTE TRIS CONTENDO EXTRATO BRUTO DE MAURITIA FLEXUOSA.

Abstract

RESUMO: Este estudo teve como objetivo avaliar os efeitos do extrato de Mauritia flexuosa em diluente Tris, sobre o sêmen caprino criopreservado. Foram utilizados dezenove ejaculados, oriundos de caprinos das raças Gurguéia e Pardo Alpina, inteiros e clinicamente saudáveis. Após coleta foi realizada a avaliação das características macro e microscópicas, seguido pela diluição com os seguintes tratamentos: G5 (solução TRIS+5% de extrato de Mauritia flexuosa), G10 (solução TRIS+ 10% de extrato de Mauritia Flexuosa) e GC (TRIS gema). Após diluição o sêmen foi envasado em palhetas francesas de 0,25 mL e criopreservado em aparelho automatizado TK 3000® em curva rápida, posteriormente, as palhetas foram imersas em nitrogênio líquido (-196°C) e, por fim, raqueadas e armazenadas em botijões criogênicos. As partidas foram descongeladas em banho-maria a 37°C por 30 segundos, acondicionadas em microtubos e homogeneizadas para a análise do TTR lento, e morfologia. A análise estatística dos dados foi realizada pelo PROC GLM (The General Linear Models Procedure) do software SAS e quando verificada significância (p<0,05) procedeu-se o teste de Tukey. No sêmen fresco, as características físicas mantiveram-se dentro dos parâmetros normais estabelecidos. Após descongelação, no TTR houve diferença significativa quanto ao grupo controle (16,05±9,06) e G10 (1,73±0,56) no parâmetro motilidade até o tempo de 60 minutos, membrana plasmática (7,42±6,86) e membrana mitocondrial (11,05±11,07). Como conclusão, observou-se que o grupo controle contendo como diluidor a gema de ovo obteve os melhores resultados comparado aos grupos contendo diferentes concentrações de Mauritia flexuosa, contudo a presença de motilidade e vigor nos grupos que contêm o citado extrato demonstra que ainda pode torna-se uma alternativa viável na criopreservação de sêmen. --------------- ABSTRACT: This study aimed to evaluate the effects of Mauritia flexuosa extract , increased by TRIS extensor on the goat semen cryopreserved. They used four goats , clinically healthy. For the collection was used only artificial vagina, totaling ninethy ejaculated. After collection was performed evaluating the macro and microscopic characteristics, followed by dilution with the following treatments: G5 (TRIS solution + 5% Mauritia flexuosa extract), G10 (TRIS solution + 10% Mauritia flexuosa extract) and GC (TRIS Yolk). After diluting semen was packaged in 0.25 ml French straws and cryopreserved in appliance automated TK 3000®, subsequently, the pallets were immersed in liquid nitrogen (-196 ° C) and finally stored and placed on racks in cryogenics tanks. The pallets were thawed in a water bath at 37 ° C for 30 seconds, put into microtubes and homogenized for analysis of slow TTR, (RTD Test) and morphology. The integrity of the membrane and mitochondrial potential were analyzed by means of fluorescent probes in epifluorescence microscope. Data were analyzed using the ANOVA. The indices obtained from motility and vigor were evaluated by PROC GLM and when checked significance (p ≥0,05) proceeded to the Tukey test. In fresh semen, the physical characteristics remained within the established normal parameters. After thawing, the TTR was no significant difference in the control group (16,05±9,06) and G10 (1,73±0,56) in motility parameter until the time of 60 minutes, the plasma membrane and mitochondrial membrane(11,05±11,07). In conclusion, it was found that the control group containing as a dilute egg yolk obtained the best results compared to groups containing different concentrations of Mauritia flexuosa, however, the presence of motility and force the groups containing the aforementioned extract shows that it can still make if a viable alternative in semen cryopreservation.
