ESTABILIDADE TEMPORAL DAS PROPRIEDADES BIOLÓGICAS DO SOLO APÓS APLICAÇÃO DE LODO DE CURTUME COMPOSTADO

RESUMO A aplicação de lodo de curtume compostado (LCC) levanta preocupações sobre seus efeitos nas propriedades biológicas do solo a longo prazo, especialmente em relação à contaminação por crômio (Cr3+). Nesse sentido, o presente estudo teve como objetivo avaliar o efeito das aplicações sucessivas de LCC na biomassa e atividade microbiana do solo, bem como a estabilidade temporal das propriedades biológicas do solo, correlacionando a atividade e biomassa microbiana do solo com os atributos químicos, especialmente os teores de Cr. Foram coletadas amostras de solo na profundidade de 0-10 cm em locais com aplicação sucessiva do LCC que possuem diferentes taxas de aplicação (2,5 Mg ha-1 - menor e 20 Mg ha-1 - maior), juntamente com um grupo controle sem aplicação (0 Mg ha-1). Foram avaliadas as atividades do Carbono da Biomassa Microbiana (CBM) e Nitrogênio da Biomassa Microbiana (NBM), respiração do solo (RS), umidade e temperatura do solo, além das enzimas desidrogenase e urease nos dias 0, 30, 60, 90 e 150 após aplicação do LCC. Os resultados mostraram que as propriedades biológicas do solo variaram conforme os tratamentos e o período de coleta, e que as variações no microclima do solo, como umidade, temperatura, pH e contaminação por Cr, tiveram um impacto significativo nessas propriedades. A análise da curva de resposta principal (PRC) revelou que o tratamento com a dose mais alta de composto apresentou uma variação maior nas propriedades microbianas do solo. Os parâmetros mais responsivos foram a desidrogenase, urease e CBM. No entanto, a estabilidade temporal das propriedades biológicas do solo foi maior na taxa mais baixa de aplicação do composto. Esses resultados indicam que a aplicação sucessiva do LCC pode afetar a biomassa e a atividade microbiana do solo, sendo que a estabilidade temporal dessas propriedades é influenciada pela taxa de aplicação do LCC, sendo a menor taxa a que proporcionou maior estabilidade.
ABSTRACT Application of composted tannery sludge (LCC) raises concerns about its long-term effects on soil biological properties, especially in relation to chromium (Cr3+) contamination. In this sense, the present study aimed to investigate the effect of successive applications of LCC on the biomass and microbial activity of the soil, as well as the temporal stability of the biological properties of the soil, correlating the activity and microbial soil biomass with the chemical attributes, especially the contents of Cr. Soil samples were collected at a depth of 0-10 cm in sites with successive application of LCC that have different application rates (2.5 Mg ha-1 - lower and 20 Mg ha-1 - higher), together with a control group without application (0 Mg ha-1). The activities of Microbial Biomass Carbon (MBC) and Microbial Biomass Nitrogen (NBM), soil respiration (RS), soil moisture and temperature, in addition to dehydrogenase and urease enzymes, were evaluated on days 0, 30, 60, 90 and 150 after applying the LCC. Results showed that soil biological properties varied according to treatments and collection period, and that variations in soil microclimate, such as moisture, temperature, pH and Cr contamination, had a significant impact on these properties. Analysis of the main response curve (PRC) revealed that the treatment with the highest dose of compost showed a greater variation in the microbial properties of the soil. The most responsive parameters were dehydrogenase, urease and CBM. However, the temporal stability of soil biological properties was higher at the lowest rate of compost application. These results indicate that the successive application of LCC can affect biomass and soil microbial activity, and the temporal stability of these properties is influenced by the LCC application rate, with the lowest rate providing greater stability.

CARACTERIZAÇÃO CITOGENÉTICA E MORFOLÓGICA DE ACESSOS DE FEIJÃOFAVA (Phaseolus lunatus L.) DO BANCO DE GERMOPLASMA DA UFPI

RESUMO
O presente trabalho teve como objetivo caracterizar citogeneticamente diferentes
acessos da espécie P. lunatus L. (feijão-fava) provenientes do Banco de
Germoplasma da Universidade Federal do Piauí (BAG-UFPI) e classificar as
sementes, dos mesmos acessos, morfologicamente quanto ao seu local de origem
(Batata, Sieva ou Grande Lima). As análises citológicas basearam-se nas técnicas
de coloração convencional com Giemsa e no bandeamento com os fluorocromos
CMA e DAPI. A caracterização das sementes foi feita com base no comprimento,
largura, espessura e peso das mesmas. Todos os acessos de feijão-fava
apresentaram um conjunto cromossômico com 2n=22, tamanho cromossômico
variando de 0,85 a 3,14 m e fórmulas cariotípicas com 11M, 10M + 1SM, 9M +
2SM, 8M + 3SM, 6M + 5SM e 7M + 4SM reforçando e corroborando com sugestões
prévias de estabilidade cariotípica. Em todos os acessos, foram encontrados blocos
de heterocromatina pericentromérica, além de pelo menos dois blocos de HC
(heterocromatina) CMA terminal correspondentes aos locais flanqueadores de
regiões organizadoras de nucléolo (RONs). As bandas de CMA apresentaram brilho
e intensidades diferentes e geraram diversas fórmulas quanto à marcação
CMA/DAPI, o que permitiu estabelecer uma relação entre o padrão de coloração e a
possível classificação das sementes. Nas sementes “Grande Lima” predominaram
as fórmulas 22CMA++/DAPI- e 22CMA/DAPI-, enquanto as sementes “Sieva” tiveram
as fórmulas 20CMA/DAPI-/2CMA++/DAPI- e 20CMA/DAPI-/2CMA+/DAPI- e “Batata”
tiveram as fórmulas 20CMA/DAPI-/2CMA+/DAPI- e 20CMA/DAPI-/2CMA++/DAPI-. A
caracterização das sementes sugere que no BAG-UFPI há representantes dos dois
grupos: andino e mesoamericano, com predominância de sementes pertencentes ao
grupo “Grande Lima” (14 acessos). Características citológicas como número,
tamanho cromossômico e padrão de bandas de HC sugere que todos os acessos
compartilham um ancestral comum. O estudo do germoplasma do feijão-fava é
importante para o conhecimento da diversidade do grupo e pode gerar informações
úteis para programas de melhoramento de P. lunatus visando à conservação dos
recursos genéticos vegetais e um maior entendimento sobre a espécie e o gênero.
ABSTRACT:
The objective of this work was to characterize cytogenetically different accesses of
the Phaseolus lunatus L. species (lima bean) from the germplasm bank of the
Federal University of Piauí (BAG - UFPI) and to classify the seeds of the same
accesses, morphologically. Cytological analyzes were based on conventional staining
techniques with Giemsa and on the banding with the fluorochromes CMA and DAPI.
The characterization of the seeds was based on the length, width, thickness and
weight. All lima bean accesses presented a chromosomal set with 2n = 22,
chromosome size ranging from 0.85 to 3,14 m and karyotypic formulas with 11M,
10M + 1SM and 9M + 2SM, reinforcing and corroborating with previous suggestions
that this group have a strong karyotypic stability. In all accesses, blocks of
pericentromeric heterochromatin were found, in addition to at least two blocks of HC
(heterochromatin) terminal CMA corresponding to NORs. The CMA bands presented
different brightness and intensity and generated several formulas from the CMA/DAPI
marking, which allowed establishing a relationship between the staining pattern and
the possible classification of the seeds. In the "Big Lima" seeds, formulas
predominated was the 22CMA++/DAPI- and 22CMA/DAPI-, while the "Sieva" seeds
had the formulas 20CMA/DAPI-/2CMA++/DAPI- and 20CMA/DAPI-/2CMA+/DAPI- and
"Potato" seeds presented the formulas 20CMA/DAPI-/2CMA+/DAPI- and
20CMA/DAPI-/2CMA++/DAPI-. The characterization of the seeds suggests that in the
BAG-UFPI there are representatives of the two groups: Andean and Mesoamerican,
according to Mackie, although with majorly seeds belonging to the "Big Lima" group
(14 accessions). Cytological features such as number, chromosome size and pattern
of HC bands suggest that all accesses share a common ancestor. The study of the
lima bean germplasm is important for the knowledge of the group diversity and can
generate useful information for breeding programs of the P. lunatus species aiming at
the conservation of the vegetal genetic resources and a greater understanding on the
species and the genus.

RESUMO: O ensaio intermediário corresponde a antepenúltima etapa do programa de
melhoramento de feijão-caupi (PMFC) da Embrapa Meio-Norte e tem a finalidade de
verificar o desempenho de novas linhagens de feijão-caupi, visando a seleção das
melhores para compor os ensaios das etapas seguintes do PMFC (Ensaio avançado
e ensaios de valor de cultivo e uso-VCU) e, por último, o lançamento de novas
cultivares superiores às existentes no mercado. Nesse estudo, objetivou-se avaliar e
selecionar linhagens de feijão-caupi da classe comercial cores, subclasses canapu e
sempre-verde, no semiárido do centro-norte piauiense com superioridade para
caracteres agronômicos, comerciais, nutricionais e qualidade de cozimento. Foram
avaliados 38 genótipos (36 linhagens e duas cultivares comerciais como testemunha)
em um ensaio intermediário conduzido no delineamento em blocos completos
casualizados, com duas repetições, no município de Ipiranga do Piauí, situado na
zona semiárida do Piauí, no Nordeste do Brasil. Os caracteres avaliados foram:
número de dias para o início da floração, comprimento de vagem, número de grãos
por vagem, peso de 100 grãos, índice de grãos, qualidade comercial do grão, teores
de proteína, ferro e zinco, porcentagem de grãos cozidos e produtividade de grãos.
Adicionalmente foram realizadas análises de Deviance e estimados parâmetros
genéticos, correlação genotípica de Pearson e o ganho com a seleção simultânea via
índice de seleção de Mulamba e Mock, utilizando-se a abordagem de modelos mistos
REML/BLUP. Observou-se diferença estatística significativa, por meio do teste de
razão verossimilhança, entre os genótipos avaliados, apenas para os caracteres
NDIF, COMPV, NGV, P100G e PROD. A herdabilidade da média dos genótipos
apresentou magnitude variando de moderada a alta. As maiores estimativas do
coeficiente de correlação genotípica entre caracteres foram observadas entre COMPV
e NGV (rg = 0,61) e entre COMPV e P100G (rg = 0,54). As linhagens 36, 5, 24, 8, 2,
23, 29, 28, 34, 6, 19, 11, 7, 20, 26, 31, 22, 18, 17, 12, 25, 15, 3, 27 e 32 são as mais
promissoras para compor o ensaio avançado do PMFC da Embrapa meio-Norte.
Destacaram-se quanto à superioridade em relação à melhor testemunha (BRS Pajeú),
as linhagens Pingo de Ouro-17-96, Bico de Ouro-17-21 e Pingo de Ouro-17-69. A
seleção simultânea de caracteres nos genótipos de feijão-caupi avaliados
proporcionou maiores ganhos genéticos reais.
ABSTRACT:The intermediate test corresponds to the antepenultimate stage of the feijão-caupi
improvement program (PMFC) of Embrapa Meio-Norte and is aimed at verifying the
performance of new lines of feijão-caupi, aiming at the selection of the best to perform
the tests of the stages Following the PMFC (Advanced testing and value cultivation
and use tests-VCU) and, finally, the launch of new cultivars superior to the existing
ones on the market. Nesse studied, aimed to assess and select feijão-caupi linhagens
of the core commercial class, subclasses canapu and semper-verde, not semi-arid
from the center-north of Piauí with superiority for agronomic, commercial, nutritional
and cooking quality characters. Foram evaluated 38 genotypes (36 strains and two
commercial cultivars as testemunha) in an intermediate trial conducted without design
in randomized complete blocks, with two repetitions, in the municipality of Ipiranga do
Piauí, located in the semi-arid zone of Piauí, in the Northeast of Brazil. The characters
supported are: number of days to the start of flowering, grain compression, number of
grains per grain, weight of 100 grams, grain index, grain commercial quality, protein,
iron and zinc theories, percentage of cooked grains and productivity of grains.
Additionally, Deviance analyzes and estimated genetic parameters, Pearson's
genotypic correlation and gain with simultaneous selection via the Mulamba and Mock
selection index were carried out, using the REML/BLUP mixed model approach.
Significant statistical differences were observed, by means of plausible reason testing,
between the validated genotypes, only for the NDIF, COMPV, NGV, P100G and PROD
characters. The average herdability of the two genotypes presented magnitude varying
from moderate to high. The highest estimates of the genotypic correlation coefficient
between foram characters observed between COMPV and NGV (rg = 0.61) and
between COMPV and P100G (rg = 0.54). As linhagens 36, 5, 24, 8, 2, 23, 29, 28, 34,
6, 19, 11, 7, 20, 26, 31, 22, 18, 17, 12, 25, 15, 3, 27e 32 are the most promising for
advanced testing of the PMFC of the Meio-Norte Embrapa. We will highlight the
superiority in relation to the best testemunha (BRS Pajeú), as the linhagens Pingo de
Ouro-17-96, Bico de Ouro-17-21 and Pingo de Ouro-1769. Simultaneous selection of
traits in tested feijão-cowpea genotypes provided greater real genetic gains.

RESUMO: A fração viva do solo é essencial para seu funcionamento, sendo a esta atribuídos muitos processos que regem a manutenção e a funcionalidade do mesmo. A rizosfera refere-se à região do solo influenciada pelas raízes, com máxima atividade microbiana. O nitrogênio (N) é um nutriente importante para as plantas e influencia o crescimento das raízes. Assim, a fertilização com este nutriente pode contribuir com o desenvolvimento da planta e, consequentemente, pode moldar a comunidade microbiana rizosférica. Este estudo levantou a hipótese de que bactérias rizosféricas, principalmente ligados à ciclagem de N, poderiam ser influenciados pela fertilização com N. O milho foi cultivado (hibrido AG1061) em tratamentos não fertilizados e N-fertilizados e a comunidade bacteriana rizosférica foi avaliada por sequenciamento de 16S rRNA. Observou-se que a comunidade bacteriana rizosférica em estudo apresentou maior riqueza nas amostras sem N, além de apresentar comunidade bacteriana distinta e predominância de Actinobacteria em comparação ao milho fertilizado com N. Bacillus aumentou na rizosfera do milho fertilizado com N, enquanto Sphingomonas, Bradyrhizobium e Rhizobium aumentaram na rizosfera não fertilizada. As bactérias generalistas superaram as especialistas na rizosfera do milho fertilizado com N. Os resultados deste estudo confirmam que o N é um nutriente importante que influencia a comunidade microbiana rizosférica do milho.
ABSTRACT: The living fraction of the soil is essential for its functioning, being attributed to many processes that govern its maintenance and functionality. The rhizosphere refers to the soil region influenced by the roots, with maximum microbial activity. Nitrogen (N) is an important nutrient for plants and influences root growth. Thus, fertilization with this nutrient can contribute to plant development and, consequently, can shape the rhizospheric microbial community. This study hypothesized that rhizospheric bacteria, mainly linked to N cycling, could be influenced by N fertilization. Maize was grown (hybrid AG1061) in unfertilized and N-fertilized treatments and the rhizospheric bacterial community was evaluated by sequencing of 16S rRNA. It was observed that the rhizospheric bacterial community under study showed greater richness in samples without N, in addition to presenting a distinct bacterial community and predominance of Actinobacteria compared to corn fertilized with N. Bacillus increased in the rhizosphere of corn fertilized with N, while Sphingomonas, Bradyrhizobium and Rhizobium increased in the unfertilized rhizosphere. Generalist bacteria outperformed specialists in the rhizosphere of N-fertilized maize. The results of this study confirm that N is an important nutrient that influences the rhizospheric microbial community of maize.
