RESUMO:
O aumento drástico da prevalência de patologias associadas ao ganho de peso é
resultado do desequilíbrio no balanço energético corporal, o que caracteriza uma
desregulação neuroendócrina e leva a um grupo heterogêneo de condições com
múltiplas causas. Frente à importância das alterações causadas por esse
desbalanço, estratégias são empregadas com a finalidade de elevar a eficácia de
perda de peso, como por exemplo, o uso de nutrientes presentes nos alimentos,
como o óleo de peixe, capaz de ligar-se a receptores importantes na manutenção da
homeostase energética, associado à estimulação do receptor β3-adrenérgico, um
dos principais responsáveis pela termogênese e gasto energético. O presente
estudo teve como objetivo avaliar se a utilização do óleo de peixe (OP) potencializa a
ação β3-adrenérgica do agonista mirabegrona (MIRA) em ratos. Após o estudo doseresposta
foram selecionadas as doses com maior efeito para os tratamentos via oral
(V.O) e intracerebroventricular (ICV), estas doses foram utilizadas no tratamento crônico
(8 dias), os animais foram segregados em quatro grupos, Salina, OP, MIRA e OP+MIRA.
A associação de OP+MIRA (V.O) mostrou-se mais eficaz que os demais tratamentos
para, redução do peso corporal, consumo de ração e aumento da temperatura
interescapular, além de aumento da expressão de genes responsáveis pelo fenótipo
termogênico (LPL, UCP1 e ADRB3) e redução da expressão do gene relacionado à
adipogênese (PPARγ) no tecido adiposo branco epididimal (TABepi) onde a frequência
de distribuição referente à área dos adipócitos foi menor para o grupo OP+MIRA. Após a
estimulação da termogênese através do frio o tratamento OP+MIRA sobrepôs seu efeito
termogênico em relação aos grupos salina e OP, e após exposição contínua ao calor o
tratamento OP+MIRA manteve a temperatura interescapular maior que os demais
tratamentos mesmo em uma condição de redução da taxa metabólica. O protocolo
realizado com um β-bloqueador revelou que apenas os grupos MIRA e OP+MIRA são
influenciados pela via simpática no aumento da temperatura interescapular, porém sem
promover aumento da pressão arterial média, verificado através do protocolo de pressão
arterial média por medida indireta. A associação entre OP+MIRA no tratamento ICV
promoveu redução no consumo de ração e a frequência de distribuição referente à área
dos adipócitos deste grupo foi menor que os demais tratamentos. Conclui-se que a
terapia combinada utilizando OP e MIRA mostra-se eficaz na modulação do balanço
energético, com efeitos sobre o peso corporal, o consumo de ração e termogênese
interescapular podendo constituir uma estratégia futura para estudos de tratamento
e prevenção da obesidade.
ABSTRACT:
The drastic increase in the prevalence of pathologies associated with weight gain is a
result of imbalance in body energy balance, which characterizes neuroendocrine
dysregulation and leads to a heterogeneous group of conditions with multiple causes.
Given the importance of the changes caused by this imbalance, strategies are
employed to increase the effectiveness of weight loss, such as the use of nutrients
present in food, such as fish oil, capable of binding to receptors. important in
maintaining energy homeostasis, associated with stimulation of the β3-adrenergic
receptor, one of the main factors responsible for thermogenesis and energy
expenditure. The present study aimed to evaluate if the use of fish oil (OP)
potentiates the β3-adrenergic action of mirabegrone agonist (MIRA) in rats. After the
dose-response study, the doses with the greatest effect for oral (VO) and
intracerebroventricular (ICV) treatments were selected. These doses were used for
chronic treatment (8 days), the animals were segregated into four groups, Salina,
OP. , MIRA and OP + MIRA. The association of OP + MIRA (VO) was more effective
than the other treatments for body weight reduction, feed intake and interscapular
temperature increase, as well as increased expression of genes responsible for
thermogenic phenotype (LPL, UCP1 and ADRB3) and reduction of adipogenesisrelated
gene (PPARγ) expression in epididymal white adipose tissue (TABepi) where
the frequency of distribution regarding adipocyte area was lower for the OP + MIRA
group. After cold thermogenesis stimulation, the OP + MIRA treatment overlapped its
thermogenic effect in relation to the saline and OP groups, and after continuous
exposure to heat, the OP + MIRA treatment kept the interscapular temperature
higher than the other treatments, even under normal conditions. reduction of
metabolic rate. The protocol performed with a β-blocker revealed that only the MIRA
and OP + MIRA groups are influenced by the sympathetic pathway in the increase of
the interscapular temperature, but without promoting an increase in mean arterial
pressure, verified through the indirect measure mean arterial pressure protocol. The
association between OP + MIRA in the ICV treatment promoted a reduction in feed
intake and the frequency of distribution regarding the adipocyte area of this group
was lower than the other treatments. It can be concluded that combined therapy
using OP and MIRA is effective in modulating energy balance, with effects on body
weight, feed intake and interscapular thermogenesis and may be a future strategy for
obesity treatment and prevention studies.

A Cistite hemorrágica é o principal efeito adverso dose-limitante do uso clínico das oxazafosforinas, incluindo a ifosfamida (IFOS). Tal evento ocorre através da formação da acroleína, metabólito responsável pela urotoxicidade desses fármacos, resultando em aumento do estresse oxidativo e produção de citocinas pró-inflamatórias, que culminam na degradação do tecido vesical. Diante disso, identifica-se uma necessidade para o desenvolvimento de um citoprotetor eficaz que possa vir a ser utilizado como adjuvante na terapia oncológica com as oxazafosforinas. Os derivados do acido gálico possuem efeitos antineoplásicos, anti-inflamatórios e antioxidantes, o que pressupõe que o galato de isoproprila (GIP) seja potencialmente útil na pesquisa de novos protótipos que possam mitigar reações inflamatórias agudas induzidas por IFOS. Neste contexto, o objetivo desta pesquisa foi analisar o efeito protetor do GIP frente à cistite hemorrágica induzida por IFOS em modelo animal. O modelo de cistite hemorrágica foi induzido por dose única do antineoplásico IFOS, precedido de pré-tratamento com salina ou GIP (6,25; 12,5; 25; 50 mg/kg, vo) em Mus musculus (CEUA 453/2017). Para analisar a redução do dano foram avaliados a massa úmida vesical (MUV), teor de hemoglobina, o extravasamento do corante azul de Evans na matriz vesical e a Proteina C reativa (PCR) por um método imunoturbidimétrico. As citocinas inflamatórias (TNF- α e IL-1β) foram dosadas por técnica de imunoensaio ELISA. Os dados de inibição foram calculados em relação ao controle negativo (CN) e as significâncias calculadas considerando p<0,05. Os resultados mostram que o pré-tratamento com o GIP (12,5 e 25 mg/kg) reduziu significativamente em 29,73% e 36,86%, respectivamente, a massa úmida vesical e atenuou de forma significativa, a hemorragia (30,1% e 54,55%, respectivamente), houve uma redução de sinais inflamatórios nas bexigas dos animais pre-tratados com GIP 25mg/kg, tanto na analise microscópica quanto macroscópica. O GIP (25 mg/kg) reduziu o significativamente o extravasamento vascular de proteínas (42,94%), os níveis de MDA (32,53%), manteve os níveis de SOD a 89,27%, de forma semelhante ao MESNA (fármaco padrão). Em relação aos níveis de GSH, o GIP 25 mg/kg, manteve o valor à 35,87% em relação ao SHAM , além de diminuir a PCR em 56,41%. Na avaliação de citocinas, o GIP na dose de 25 mg/kg foi capaz de diminuir os níveis de TNF-α (88,77 %) e IL-1β 62,87%). Com base no exposto, pode-se concluir que o GIP foi capaz de atenuar os sinais da cistite hemorrágica induzida por ifosfamida em camundongos e que representa uma promissora opção terapêutica na modulação dessa condição patológica.
ABSTRACT:
Hemorrhagic cystitis is the main dose-limiting adverse effect of clinical use of oxazaphosphorins, including ifosfamide (IFOS). This event occurs through the formation of acrolein, metabolite responsible for the urotoxicity of these drugs, resulting in increased oxidative stress and production of proinflammatory cytokines, which culminate in the degradation of the bladder tissue. In view of this, a need is identified for the development of an effective cytoprotector that may be used as an adjunct in oncological therapy with oxazaphosphorins. Gallic acid derivatives have antineoplastic, anti-inflammatory and antioxidant effects, which presupposes that isopropyl gallate (IPG) is potentially useful in the search for new prototypes that can mitigate IFOS-induced acute inflammatory reactions. In this context, the objective of this research was to analyze the protective effect of IPG against IFOS-induced hemorrhagic cystitis in an animal model. The hemorrhagic cystitis model was induced by a single dose of IFOS antineoplastic, preceded by saline or IPG pretreatment (6.25; 12.5; 25; 50 mg / kg, vo) in Mus musculus (CEUA 453/2017). In order to analyze the damage reduction, bladder wet mass (BWM), hemoglobin content, Evans blue dye leakage in the bladder matrix and C-reactive protein (CRP) were evaluated by an immunoturbidimetric method. Inflammatory cytokines (TNF-α and IL-1β) were measured by ELISA immunoassay. Inhibition data were calculated in relation to the negative control (NC) and the significance calculated considering p <0.05. Results show that pretreatment with IPG(12.5 and 25 mg / kg) significantly reduced bladder wet mass by 29.73% and 36.86%, respectively, and significantly attenuated bleeding (30,1% and 54.55%, respectively), there was a reduction of inflammatory signs in the bladders of animals pre-treated with IPG 25mg / kg, both in the microscopic and macroscopic analysis. IPG (25 mg / kg) significantly reduced vascular protein extravasation (42.94%), MDA levels (32.53%), maintained SOD levels at 89.27%, similar to MESNA (standard drug). Regarding GSH levels, IPG 25 mg / kg maintained the value at 35.87% compared to SHAM, besides reducing CRP by 56.41%. In the cytokine evaluation, 25 mg / kg IPG was able to decrease the levels of TNF-α (88.77%) and IL-1β 62.87%). Based on the above, it can be concluded that IPG was able to attenuate the signs of ifosfamide-induced hemorrhagic cystitis in mice and represents a promising therapeutic option in modulating this pathological condition.

RESUMO:O bacuri (Platonia insignis Mart.) tem sido utilizado na medicina popular por suas atividades anti-inflamatórias e cicatrizante, mas não há relatos de efeito hipocolesterolemiante e, por isso, foi avaliado os efeitos da manteiga da semente do bacuri (MB) sobre o perfil lipídico, estado antioxidante, função hepática, peso dos órgãos e parâmetros metabólicos em hamsters. Além disso, foi avaliada a toxicidade aguda em ratos e a atividade antioxidante in vitro. Hamsters machos, recém-desmamados, receberam ração contendo caseína (20%), gordura de coco (13,5%) e colesterol (0,1%) durante 21 dias para indução da dislipidemia. O grupo controle normal (CN, n=8) recebeu ração padrão. Após indução, os animais com dislipidemia foram aleatoriamente separados em três grupos, todos mantidos com ração hiperlipídica durante os 28 dias de tratamento: controle dislipidemia (CD, n=8); MB 25 mg/kg/dia (MB-25, n=10); MB 50 mg/kg/dia (MB-50, n=10). Ao final do tratamento foram determinadas as concentrações séricas de triglicérides (TG), colesterol total (CT), LDL colesterol (LDL-c), HDL-c, malondialdeído (MDA), alanina aminotransferase (ALT), aspartato aminotransferase (AST) e fosfatase alcalina (FAL), superóxido dismutase (SOD) em eritrócitos, peso do fígado, coração e gordura retroperitoneal. Durante todo o período experimental, acompanhou-se a ingestão alimentar, ganho de peso e crescimento nasoanal dos grupos tratados e não tratados, e, ao final do período experimental, calculou-se os índices de massa corpórea e de Lee dos grupos. Para a toxicidade aguda, ratos foram divididos em dois grupos: grupo controle (tween 80 em água destilada) e grupo MB 2.000 mg/kg. Atividade antioxidante avaliada pelo método do radical livre 2,2-difenil-1-picrilhidrazil (DPPH). A comparação entre grupos foi realizada por análise de variância e teste de Tukey. A pesquisa foi aprovada por Comissão de Ética em Uso de Animais (CEUA/UFPI 197/16). Não houve diferenças estatisticamente significativas entre os grupos MB-25 e MB-50 com CN e CD em relação a TG e CT. Contudo, MB-25 apresentou HDL-C significativamente maior (51,40 ± 1,69) (p<0,05) e LDL-c menor (103,80 ± 6,87) (p<0,05), bem como MB-50 apresentou nível maior (p<0,05) de HDL-c (51,00 ± 2,34) e menor (p<0,05) de LDLc (100,50 ± 3,95) comparados a CD (LDLc=132,70 ± 9,41; HLDc= 41,00 ± 2,94). Além disso, CD apresentou níveis de TG (88,00 ± 4,30), COL (183,00 ± 9,00), HDL-c (41,00 ± 2,94) e LDL-c (132,70 ± 9,41) maiores (p<0,05) que CN (TG= 35,00 ± 5,00; CT= 95,00 ± 3,50; HDLc= 26,88 ± 2,15; LDLc= 60,60 ± 2,26). Não foram observadas diferenças (p>0,05) em relação à: MDA, ALT, AST, FAL, peso do coração e da gordura retroperitoneal, ingestão alimentar, ganho de peso, crescimento nasoanal e massa corpórea. Entretanto, observou-se um aumento isolado no peso do fígado dos animais dos grupos MB-25 (5,32 ± 0,30) e MB-50 (5,23 ± 0,32), em relação aos do controle normal (4,07 ± 0,17). A MB apresentou baixa toxicidade aguda e demonstrou potencial antioxidante in vitro (EC50=493,10 μg/mL). Quanto aos níveis de SOD, não foram observadas diferenças entre MB-25 (1.557 ± 53,93), CN (1.437 ± 71,70) e CD (1.430 ± 128,90), entretanto, MB-50 (1.868 ± 131,80) apresentou níveis significativamente maiores (p<0,05) que CN e CD. Em conclusão, o estudo demonstrou que a MB, nas doses e tempo de tratamento empregados, apresenta repercussões positivas sobre o perfil lipídico em hamsters hipercolesterolêmicos. A maior dose de 50 mg/kg produziu aumento isolado nos níveis de SOD, indicando atividade antioxidante e não apresentou toxicidade sistêmica. A MB não demonstrou toxicidade aguda e possui potencial antioxidante in vitro.
ABSTRACT:Bacuri (Platonia insignis Mart.) has been used in folk medicine for its anti-inflammatory and healing effects, but there are no reports of hypocholesterolemic effect and for this reason, we evaluated the effects of bacuri seed butter (MB) on blood lipid profile, antioxidant status, hepatic function and metabolic parameters in hamsters with diet-induced hypercholesterolemia. In addition, acute toxicity in rats and in vitro antioxidant activity were evaluated. Freshly weaned male hamsters were given chow containing casein (20%), coconut oil (13.5%) and cholesterol (0.1%) for 21 days to induce dyslipidemia. The normal control group (CN, n=8) received standard chow. After induction, the animals with dyslipidemia were randomly separated into three groups, all of them maintained with hyperlipidic chow during 28 days of treatment: Dyslipidemia control (CD, n=8); MB 25 mg/kg/d (MB-25, n=9) and 50 mg/kg/d (MB-50, n=9). At the end, the serum levels of triglycerides (TG), total cholesterol (TC), LDL-cholesterol (LDLc), HDL-cholesterol (HDLc), malondialdehyde (MDA), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (FA), the erythrocytes levels of superoxide dismutase activity (SOD), and liver, heart and retroperitoneal adipose tissue weights were measured. Statistical analysis was performed using one-way ANOVA, followed by Tukey’s multiple comparisons test. The research was approved by the Committee on Ethics in Animal Use (CEUA/UFPI 197/16). There were no statistically significant differences between the groups MB-25 and MB-50 with CN and CD in relation to TG, COL. However MB-25 presented higher (p<0.05) HDLc (51.4 ± 1.69) and lower (p<0.05) LDLc (103.8 ± 6.87) levels, as well as MB-50 presented higher (p<0.05) HDLc (51.00 ± 2.34) and lower LDLc (100.5 ± 3.95) levels, compared to CD (LDLc=132.7 ± 9.41; HLDc= 41.00 ± 2.94). Furthermore, CD has shown TG (88.0 ± 4.3), COL (183.00 ± 9.0), HDLc (41.00 ± 2.94) and LDLc (132.7 ± 9.41) levels higher (p<0.001) than CN (TG= 35.00 ± 5,0; CT= 95.00 ± 3.5; HDLc= 26.88 ± 2.15; LDLc= 60.60 ± 2.26). No differences were found (p>0.05) in relation to MDA, ALT, AST, FA, food intake, weight gain, nasoanal growth, body mass index, and heart and adipose tissue weights. However, MB (25 and 50 mg/kg/day) produced a significant (p<0,05) increase in liver weight (5,32 ± 0,30; 5,23 ± 0,32, respectively), when compared to the normal control group. MB has shown antioxidant activity in vitro (EC50=493.10 μg/mL) and no signs of acute toxicity in vivo. There was no difference between MB-25 (1.557.24 ± 132.10), CN (1.436.6 ± 175.6) and CD (1.430.0 ± 257.8) in relation to SOD, but it was increased (p<0.05) in MB-50 (1.868 ± 131.80) compared to CN and CD. Thus, the present study demonstrated that MB had a positive impact on the serum lipid profiles in hypercholesterolemic hamsters. The higher dose of 50 mg/kg produced isolated increase in SOD levels, indicating antioxidant activity, and showed no systemic toxicity. Moreover, MB presented antioxidant activity in vitro and no acute toxicity in vivo.

RESUMO:Os glicocorticóides quando administrados em tratamento prolongado (> 3 semanas) ou em excesso no organismo, podem levar ao desenvolvimento da Síndrome de Cushing (SC), que é caracterizada por múltiplos distúrbios metabolicos. Pesquisas com suplementação com aminoácidos de cadeia ramificada (ACR), especialmente a leucina (LEU), demonstram que este nutriente é capaz de melhorar a saúde metabólica em animais obesos. No entanto, evidencias recente aprontam que a ausência dos ACR ou da LEU na dieta são capazes de melhorar alguns aspectos metabólicos, por reduzir a adiposidade e aumentar a termogênese. Portanto, não está claro se a suplementação dietética com LEU seria uma estratégia benéfica. Para ajudar a resolver essa questão, examinamos o efeito da suplementação com LEU nos distúrbios metabólicos atribuídos à SC. Para tanto, usamos o tratamento nutricional de longo prazo com LEU no modelo farmacológico de SC, induzido por excesso de dexametasona (DEXA), (dose 1,0mg/kg) aplicado quatro vezes semanais via intraperitoneal. Descobrimos que a suplementação com LEU piora vários dos quadros metabólicos que caracterizam a SC como a adiposidade, a hipertriglicidemia, hipercolesterolemia e hiperalbuminemia. Bem como, favorece maior acúmulo de gordura e marcadores de lesão hepática. Ademais, verificamos que este aminoácido não apresentou potencial antioxidante, redução na peroxidação lipídica e aumento da atividade de enzimas antioxidantes. Além disso, nossos resultados indicam que LEU piora a saúde metabólica em animais com SC, e sugerem que o aumento específico de LEU alimentar pode representar uma estratégia ariscada no modelo experimental estudado.
ABSTRACT:Glucocorticoids when given in prolonged (> 3 weeks) or in excess in the body may lead to the development of Cushing's Syndrome (SC), which is characterized by multiple metabolic disorders. Research with branched chain amino acid (ACR) supplementation, especially leucine (LEU), demonstrates that this nutrient is capable of improving metabolic health in obese animals. However, recent evidence indicates that the absence of ACR or LEU in the diet are able to improve some metabolic aspects, by reducing adiposity and increasing thermogenesis. Therefore, it is not clear whether dietary supplementation with LEU would be a beneficial strategy. To help address this issue, we examined the effect of LEU supplementation on metabolic disorders attributed to SC. For this, we used the long-term nutritional treatment with LEU in the pharmacological model of SC, induced by excess dexamethasone (DEXA), (dose 1.0mg / kg) administered four times weekly intraperitoneally. We found that supplementation with LEU worsens several of the metabolic conditions that characterize CS such as adiposity, hypertriglyceridemia, hypercholesterolemia and hyperalbuminemia. As well as, it favors greater accumulation of fat and markers of liver damage. In addition, surprisingly, we verified that this amino acid presented antioxidant potential, reduction in lipid peroxidation, even when it presents high myeloperoxidase activity. Our results indicate that LEU worsens metabolic health in SC animals, and suggests that the specific increase in dietary LEU may represent a strategy in the experimental model studied.
