RESUMO
Ephedranthus pisocarpus R. E. Fries (Annonaceae) é conhecida como envira de cocho ou conduru. Este trabalho relata os constituintes químicos isolados e identificados no extrato etanólico das folhas de E. pisocarpus, a análise por ESI-ITMS da fração acetato de etila e a investigação das atividades biológicas do extrato etanólico e frações. O estudo fitoquímico das frações hexano e diclorometano da partição do extrato etanólico das folhas de E. pisocarpus resultou no isolamento do óxido de cariofileno (1), espatulenol (2), uma mistura contendo dois sesquiterpenoides: 4α,10β-aromadendranodiol (3) e 1α-hidróxi-espatulenol (4), T-muurolol (5), 1β,6α-di-hidróxi-4(15)-eudesmeno (6), 24-metilenocicloartanol (7), 24-metilenocicloartano-3β,21-diol (8) e uma mistura dos esteroides: colesterol (9), campesterol (10), campestanol (11), estigmasterol (12), Δ22-estigmastenol (13), sitosterol (14) e sitostanol (15). As estruturas dos compostos isolados foram identificadas por RMN 1H e 13C, CG-EM e por comparação com dados da literatura. Na fração acetato de etila foram identificados cinco compostos por ESI-ITMS: ácido quínico (16, m/z 191) vitexina (17, m/z 431), sulfoquinovosilmonoacilglicerol (18, m/z 555), canferol-3-O-rutinosídeo (19, m/z 593) e rutina (20, m/z 609). Na avaliação da atividade antioxidante pelo método com o DPPH, a fração acetato de etila apresentou percentual de atividade antioxidante de 22,55% na maior concentração testada (250 μg mL-1), correlacionando com o maior teor de fenóis totais (155,22±3,04 EAG g-1 amostra). O extrato etanólico e frações apresentaram atividade fotoprotetora, sendo as frações acetato de etila (FPS 3,90) e aquosa (FPS 3,85) as mais ativas. No ensaio de citotoxicidade frente à linhagem B16-F10 o extrato EtOH e frações hexano, diclorometano e acetato de etila apresentaram considerável redução da viabilidade celular, sendo a fração diclorometano a mais ativa. O extrato etanólico apresentou moderada atividade antileishmania com CI50 de 82,0 μg mL-1. As frações hexano (CI50= 45,1 μg mL-1) e diclorometano (CI50= 23,3 μg mL-1) foram as mais ativas frente as promastigotas de Leishmania amazonensis. Este é o primeiro relato de investigação destas atividades biológicas em E. pisocarpus, bem como da ocorrência de todos esses metabólitos no gênero Ephedranthus e dos compostos 4 e 18 na família Annonaceae.

Ephedranthus pisocarpus R. E. Fries (Annonaceae) is known as envira de cocho or conduru. This work reports the chemical constituents isolated and identified in the ethanolic extract of the leaves of E. pisocarpus, the analysis by ESI-ITMS of the ethyl acetate fraction and the investigation of the biological activities of the ethanolic extract and fractions. The phytochemical study of the hexane and dichloromethane fractions of the ethanolic extract of E. pisocarpus leaves resulted in the isolation of caryophyllene oxide (1), spathulenol (2), a mixture containing two sesquiterpenoids: aromadendrane-4α, 10β-diol (3) and 1α-hidroxy-spathulenol (4), T-muurolol (5), 1β,6α-di-hydroxy-4(15)-eudesmene (6), 24-methylenecycloartanol (7), 24-methylenecycloartane-3β,21-diol (8) and a mixture of steroids: cholesterol (9), campesterol (10), campestanol (11), stigmasterol (12), Δ22-stigmastenol(13), sitosterol (14) and sitostanol (15). Structures of the isolated compounds were identified by 1H and 13C NMR, GC-MS and by comparison to the literature. In the ethyl acetate fraction five compounds were identified by ESI-ITMS: quinic acid (16, m/z 191), vitexin (17, m/z 431), sulfoquinovosylmonoacylglycerol (18, m/z 555), kaempferol-3-O-rutinoside (19, m/z 593) and rutin (20, m/z 609). In the evaluation of the antioxidant activity by the DPPH method, the ethyl acetate fraction presented a percentage of antioxidant activity of 22.55% in the highest concentration tested (250 μg mL -1), correlating with the highest total phenol content (155.22 ± 3.04 EAG g-1 sample). The ethanolic extract and fractions showed photoprotective activity, with the ethyl acetate (SPF 3,90) and aqueous (SPF 3,85) fractions being the most active. In the cytotoxicity assay against the B16-F10 line the EtOH extract and hexane, dichloromethane and ethyl acetate fractions showed a considerable reduction of cell viability, with the dichloromethane fraction being the most active. The ethanolic extract presented moderate antileishmanial activity with an IC50 of 82.0 μg mL-1. The hexane (IC50= 45.1 μg mL-1) and dichloromethane (IC50= 23.3 μg mL-1) fractions were the most active against Leishmania amazonensis promastigotes. This is the first report of investigation of these biological activities in E. pisocarpus, as well as the occurrence of all these metabolites in the genus Ephedranthus and of compounds 4 and 18 in the Annonaceae family. |
