RESUMO Com o avanço das biotécnicas reprodutivas e do melhoramento animal, em todas as espécies de interesse econômico, o aprimoramento das técnicas de conservação de germoplasma se tornaram cada vez mais necessárias. A criopreservação de sêmen é uma dessas principais técnicas de conservação, no entanto, elas geram danos irreversíveis nas células espermáticas, a exemplo da lipoperoxidação, causada por espécies reativas ao oxigênio (EROs). Diante disso, se faz necessário o estudo de métodos que atenuem esses danos e aumentem a qualidade final do material criopreservado. Dessa forma, objetivou-se avaliar o efeito do estresse subletal, por meio da suplementação com Diazinon (120 μM), assim como o efeito dos agentes antioxidantes Trolox (75 μM), Genisteína (25 μM) e Resveratrol (20 μM), no diluidor TRIS-gema, sobre a qualidade do sêmen criopreservado de ovinos. Para tanto, foram coletadas amostras de sêmen de seis carneiros da raça Dorper clinicamente saudáveis e com os parâmetros andrológicos/seminais normais. O sêmen foi diluído em diluidor Tris-Gema com glicerol e os respectivos tratamentos, em cada experimento. Experimento 1: Controle (Tris-Gema), T1 (Tris-Gema + 120 µM de Diazinon); T2 (Tris-Gema + 75 µM de Trolox). Experimento 2: Controle (Tris-Gema); T1 (Tris-Gema + 25 µM de Genisteína); T2 (Tris-Gema + 20 µM de Resveratrol). Posteriormente, foram realizadas as análises macro e microscópicas do ejaculado pré e pós-diluição. As amostras foram criopreservadas pelo método automatizado, em curva de congelação lenta para a espécie ovina e, após 15 dias, foram realizadas as seguintes análises pós-descongelamento: análise física, avaliação da longevidade espermática pelo teste de termorresistência (TTR), avaliação morfológica pós-descongelação, avaliação da ultraestrutura espermática: integridade da membrana plasmática, integridade acrossomal, atividade mitocondrial e funcionalidade da membrana plasmática espermática. Foi utilizado no estudo o delineamento inteiramente casualizado (DIC), os dados foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey e Duncan, com um nível de significância de 5%. Observou-se um aumento significativo do vigor espermático no tratamento com Resveratrol, quando comparado ao sêmen pré-congelação (p<0,05). Os tratamentos não geraram alterações nos parâmetros da ultraestrutura espermática (p>0,05). Não houve diferença entre os tratamentos na avaliação da longevidade espermática pelo TTR, no entanto, houve estabilizações na motilidade e vigor espermáticos entre os tempos do TTR (p<0,05). Não houve diferença entre os tratamentos na análise de morfologia espermática do sêmen criopreservado. A funcionalidade da membrana plasmática foi reduzida nos grupos com 120 µM de Diazinon e 25 µM de Genisteína adicionadas meio diluidor. Não houve diferença entre os tratamentos na avaliação dos parâmetros cinéticos pelo CASA (p>0,05), com exceção da motilidade progressiva do T1 (Tris-Gema + 120 µM de Diazinon) que se mostrou reduzida em relação ao controle (p<0,05). Em conclusão, a suplementação de 120 μM de Diazinon ao meio diluidor Tris-Gema, não melhorou a cinética espermática bem como a qualidade estrutural dos espermatozoides criopreservados e a adição de 75 µM de Trolox manteve a qualidade espermática. A adição de 25 µM de Genisteína, ao diluidor Tris-Gema, mantém os parâmetros cinéticos espermáticos em relação ao controle, porém a suplementação de 20 µM de Resveratrol ao diluidor foi capaz de melhorar o vigor espermático do sêmen ovino criopreservado.
ABSTRACT With the advancement of reproductive biotechniques and animal improvement, in all species of economic interest, the improvement of germplasm conservation techniques has become increasingly necessary. Semen cryopreservation is one of these main conservation techniques, however, they generate irreversible damage to sperm cells, such as lipoperoxidation, caused by reactive oxygen species (ROS). Therefore, it is necessary to study methods that mitigate these damages and increase the final quality of the cryopreserved material. Thus, the objective was to evaluate the effect of sublethal stress, through supplementation with Diazinon (120 μM), as well as the effect of the antioxidant agents Trolox (75 μM), Genistein (25 μM) and Resveratrol (20 μM), on TRIS-yolk extender on the quality of cryopreserved ram semen. For this purpose, semen samples were collected from six clinically healthy Dorper rams with normal andrological/seminal parameters. The semen was diluted in Tris-Gema extender with glycerol and the respective treatments, in each experiment. Experiment 1: Control (Tris-Gem), T1 (Tris-Gem + 120 µM Diazinon); T2 (Tris-Gem + 75 µM Trolox). Experiment 2: Control (Tris-Gema); T1 (Tris-Gem + 25 µM Genistein); T2 (Tris-Gem + 20 µM Resveratrol). Subsequently, macro and microscopic analyzes of the pre- and post-dilution ejaculate were carried out. The samples were cryopreserved by the automated method, in a slow freezing curve for the ovine species and, after 15 days, the following post-thawing analyzes were carried out: physical analysis, evaluation of sperm longevity by the thermoresistance test (TTR), post morphological evaluation -thawing, evaluation of sperm ultrastructure: plasma membrane integrity, acrosomal integrity, mitochondrial activity and sperm plasma membrane functionality. The study used a completely randomized design (DIC), the data were submitted to analysis of variance and the averages compared by the Tukey and Duncan test, with a significance level of 5%. There was a significant increase in sperm vigor in the treatment with Resveratrol, when compared to pre-freezing semen (p<0.05). Treatments did not change sperm ultrastructure parameters (p>0.05). There was no difference between treatments in the evaluation of sperm longevity by TTR, however, there were stabilizations in sperm motility and vigor between TTR times (p<0.05). There was no difference between treatments in sperm morphology analysis of cryopreserved semen. Plasma membrane functionality was reduced in the groups with 120 µM Diazinon and 25 µM Genistein added to extender medium. There was no difference between treatments in the evaluation of kinetic parameters by CASA (p>0.05), with the exception of the progressive motility of T1 (Tris-Gem + 120 µM of Diazinon) which was reduced in relation to the control (p<0 .05). In conclusion, the supplementation of 120 μM of Diazinon to the Tris-Gema extender medium, did not improve the sperm kinetics as well as the structural quality of the cryopreserved spermatozoa and the addition of 75 µM of Trolox maintained the sperm quality. The addition of 25 µM of Genistein, to the extender Tris-Gema, maintains the sperm kinetic parameters in relation to the control, however the supplementation of 20 µM of Resveratrol to the extender was able to improve the sperm vigor of cryopreserved ram semen.

RESUMO GERAL
As galinhas nativas descendem de animais trazidos pelos portugueses durante o período de
colonização. Aqui sofreram processos de seleção natural durante anos, o que levou a mudanças
morfológicas e fisiológicas como mecanismos de adaptação às condições ambientais adversas.
Embora adaptadas, estas aves são menos produtivas que as linhagens de galinhas selecionadas e tem
sido substituídas gradativamente por animais melhorados, o que levou a rápida redução e/ou
diluição desses recursos genéticos. Muitas das galinhas nativas se encontram em reduzidos núcleos
de criação com mínimo controle zootécnico, na agricultura familiar, constituindo ainda a base da
subsistência para essa. A conservação e utilização sustentável destes recursos genéticos, por meio
dos programas de conservação das espécies domésticas e incentivo para a multiplicação do uso e
reconhecimento destas raças são urgentes. Objetivou-se, portanto, caracterizar fenotipicamente e
também, avaliar as características físicas dos ovos de galinhas Suras. Na caracterização fenotípica,
os caracteres quantitativos foram submetidos a uma análise de variância. A análise estatística foi
feita utilizando o método dos quadrados mínimos tendo sido realizadas análise da média, mínimo,
máximo e coeficiente de variação de cada variável e para cada núcleo. Para dos ovos, analisaram-se
diversos parâmetros físicos e dos seus componentes. Depois de recolher os resultados dos testes
acima referidos, foram realizadas a determinação do peso do albúmen por diferença entre o peso do
ovo e pesos da casca e gema, determinação percentual dos diferentes constituintes do ovo, shape
index (índice de forma) e shell index (índice de casca). A análise estatística foi efetuada recorrendo
ao programa SAS (SAS Institute, 2012). Realizou-se uma análise de variância, comparando as
médias de cada parâmetro estudado pelo teste de Tukey, através dos procedimentos General Linear
Models do programa SAS. As galinhas Suras apresentam - se fenotipicamente estruturada, com
padrões uniformes, sendo sua principal característica a ausência do pigóstilo. No que se refere aos
ovos de Suras verificou-se que possuem variabilidade nas diferentes nas características físicas que
traduzem a qualidade do ovo nas duas classes de peso (grandes e pequenos). Também se verificou
que as galinhas Suras produzem ovos mais arredondados, e com casca castanha e gema mais
alaranjada nas duas classes de pesos.
ABSTRACT GERAL
Native chickens descend from animals brought by the Portuguese during the colonization period.
Here they have undergone natural selection processes for years, which led to morphological and
physiological changes as adaptation mechanisms to adverse environmental conditions. Although
adapted, these birds are less productive than selected chicken lines and have been gradually
replaced by improved animals, which has led to the rapid reduction and/or dilution of these genetic
resources. Many of the native chickens are found in small breeding centers with minimal
zootechnical control, in family farming, still constituting the basis of subsistence for this. The
conservation and sustainable use of these genetic resources, through domestic species conservation
programs and incentives for the multiplication of use and recognition of these breeds, are urgent.
The objective, therefore, was to characterize phenotypically and also evaluate the physical
characteristics of eggs from Suras chickens. In the phenotypic characterization, the quantitative
characters were subjected to an analysis of variance. Statistical analysis was carried out using the
least squares method, with analysis of the mean, minimum, maximum and coefficient of variation
of each variable and for each nucleus. For eggs, several physical parameters and their components
were analyzed. After collecting the results of the tests mentioned above, the weight of the albumen
was determined by the difference between the weight of the egg and the weights of the shell and
yolk, percentage determination of the different constituents of the egg, shape index and shell index
(shell index). Statistical analysis was carried out using the SAS program (SAS Institute, 2012). An
analysis of variance was carried out, comparing the means of each parameter studied using the
Tukey test, using the General Linear Models procedures of the SAS program. Suras chickens are
phenotypically structured, with uniform patterns, their main characteristic being the absence of the
pygostyle. With regard to Suras eggs, it was found that they have variability in the different physical
characteristics that reflect the quality of the egg in the two weight classes (large and small). It was
also found that Suras hens produce more rounded eggs, with brown shells and more orange yolks in
both weight classes.

RESUMO – Objetivou-se definir níveis de cálcio que otimize desempenho e qualidade dos ovos, bem como as principais atividades comportamentais e fisiológicas de galinhas caipiras Canela-Preta, alimentadas com rações contendo diferentes níveis de inclusão de cálcio (2,3%, 3,0%, 3,7% e 4,4%), no período de 28 a 36 semanas de idade, sob pastejo de capim batatais (Paspalum notatum). O nível de 2,3% de cálcio atende às exigências de desempenho de postura das galinhas Canela-Preta, na fase de 28 a 36 semanas de vida das aves, tanto para dúzia como para massa de ovos. Os níveis de cálcio da ração não interferiram no comprimento, gravidade específica, espessura e resistência à quebra da casca, índice de forma e unidade Haugh do ovo, porém, o percentual da gema decresce até o nível 4,17% e o percentual da casca do ovo a partir de 3,03% de inclusão de cálcio nas dietas. Os parâmetros ósseos não foram afetados até a inclusão de 4,4% de cálcio nas dietas de galinha Canela-Preta, no período de 28 a 32 semanas de vida. Os níveis de cálcio das dietas não influenciaram os parâmetros fisiológicos o nem os comportamentos das galinhas Canela-Preta, no entanto, as aves utilizam em torno de 45% do tempo em ócio, independentemente do nível de cálcio das dietas. As galinhas caipiras Canela-preta expressam comportamentos de conforto, ingestivo e social, característicos de animais criados em sistemas com acesso a área de pastejo, independente do cálcio da dieta.
ABSTRACT - The objective was to define calcium levels that optimize performance and egg quality, as well as the main behavioral and physiological activities of Canela-Preta free-range chickens, fed diets containing different levels of calcium inclusion (2.3%, 3.0%, 3.7% and 4.4%), from 28 to 36 weeks of age, under potato grass (Paspalum notatum) grazing. The level of 2.3% calcium meets the laying performance requirements of Canela-Preta hens, in the 28 to 36 weeks of life, both for dozen and for egg mass. The levels of calcium in the diet did not interfere with the length, specific gravity, thickness and resistance to breakage of the shell, shape index and Haugh unit of the egg, however, the percentage of the yolk decreases to the level of 4.17% and the percentage of the shell egg from 3.03% inclusion of calcium in diets. Bone parameters were not affected until the inclusion of 4.4% of calcium in Canela-Preta chicken diets, in the period from 28 to 32 weeks of life. The levels of calcium in the diets did not influence the physiological parameters or the behaviors of Canela-Black hens, however, the birds spend around 45% of their time idling, regardless of the level of dietary calcium. Canela-preta free-range chickens’ express comfort, ingestive and social behaviors, characteristic of animals raised in systems with access to grazing area, regardless of dietary calcium.

RESUMO Objetivou-se estimar parâmetros genéticos, predizer valores genéticos e identificar regiões cromossômicas para resistência a endoparasitas em caprinos da raça Anglonubiana, em um modelo de repetibilidade em análise unicaracterística, com inclusão da informação genômica. As acurácias de predição dos valores genéticos foram analisadas para os dois métodos utilizados, BLUP e ssGBLUP. Além disso, investigou-se a contribuição de diferentes vias de seleção, particularmente animais genotipados e não genotipados e machos e fêmeas, para as tendências genéticas de características de resistência a endoparasitas. As características indicadoras de resistência a endoparasitas utilizadas no estudo foram escore de condição corporal (ECC), pontuação FAMACHA, ovos por grama de fezes (OPG), tratada com a escalas de transformações logarítmicas: L1OPG = log (OPG + 1) e resistência a verminose (RV) de 1.500 caprinos. Deste total, 89 animais foram genotipados utilizando o Bead ChipGoatsSNP50 da Illumina que contêm 53.347 SNPs (Single Nucleotide Polymorphism). Não houve grandes mudanças nas estimativas dos componentes de variância, nas herdabilidades e repetibilidades com a inclusão da informação genômica. O uso de informações genômicas melhorou a capacidade preditiva dos modelos, proporcionando ganhos em acurácia de até 50% para a característica OPG. Na análise das tendências genéticas, observou-se que com o uso de ssGBLUP os ganhos genéticos anuais foram significativos FAMACHA (b = -0,0022; p = 0,03), L1OPG (b = -0,0013; p = 0,02). Maior contribuição para as tendências genéticas foi observada para animais genotipados em relação aos não-genotipados, assim como maior participação das fêmeas para o ganho genético. Foram identificadas 12, 11, 12, e 16 regiões candidatas, respectivamente, para ECC, FAMACHA, L1OPG, e RV, que explicaram no mínimo 1% da variância genética em 23, do total de 30, cromossomos associados com as características em estudo. Nessas regiões foram encontrados genes conhecidamente associados a processos do sistema imunológico. Assim como genes com funções ainda desconhecidas. Os resultados indicaram que a inclusão de informações genômicas será favorável para a avaliação genética de caprinos Anglonubianos para características indicadoras de resistência parasitaria. As regiões genômicas identificadas poderão auxiliar no entendimento da arquitetura genética das características em estudo e contribuir para a seleção de caprinos com maior resistência a endoparasitas.
ABSTRACT The objective of this study was to estimate the variance components and genetic parameters, predict genetic values and identify chromosomal regions for resistance to endoparasites in Anglo-Nubian goats, in a model of repeatability in unicharacter analysis, with inclusion of genomic information. The accuracies of prediction of genetic values were analyzed for the two methods used, BLUP and ssGBLUP. Additionally, the contribution of different selection pathways, particularly genotyped and non-genotyped animals and males and females, to the genetic trends of resistance to endoparasites characteristics were investigated. The indicator characteristics of resistance to endoparasites used in the study were body condition score (ECC), FAMACHA score, eggs per gram of feces (OPG), treated with the logarithmic transformation scales: L1OPG = log (OPG + 1) and resistance to verminose (RV) of 1,500 goats were used as characteristics associated with resistance to verminose. Of this total, 89 animals were genotyped using the Illumina Bead ChipGoatsSNP50 which contains 53,347 SNPs (Single Nucleotide Polymorphism). There were no major changes in the estimates of the variance components, in the heritabilities and repeatability with the inclusion of genomic information. The use of genomic information improved the predictive capacity of the models, providing gains in accuracy of up to 50% for the OPG characteristic. In the analysis of genetic trends, it was observed that with the use of ssGBLUP the annual genetic gains were significant FAMACHA (b = -0,0022; p = 0,03), L1OPG (b = -0,0013; p = 0,02). Greater contribution to the genetic trends was observed for genotyped animals in relation to non-genotyped animals, as well as greater participation of females for the genetic gain. Twelve, eleven, twelve, and sixteen candidate regions were identified, respectively, for ECC, FAMACHA, L1OPG, and RV, which explained at least 1% of the genetic variance in 23, out of a total of 30, chromosomes associated with the characteristics under study. In these regions, genes known to be associated with immune system processes were found. As well as genes with still unknown functions. The results indicated that the inclusion of genomic information will be favorable for the genetic evaluation of Anglo-Nubian goats for indicator characteristics of parasitic resistance. The identified genomic regions could help in the understanding of the genetic architecture of the characteristics under study and contribute to the selection of goats with greater resistance to endoparasites.
